菲立磁标记胚胎干细胞移植肝损伤小鼠体内示踪研究

摘    要:目的 观察菲立磁标记胚胎干细胞肝损伤模型移植后在体内迁徙、定居、增殖及与损伤肝脏整合.方法 将129/SvJ小鼠随机分为肝损伤和无肝脏损伤组,将最适菲立磁标记细胞数量经肝脏包膜下途径移植,通过对移植后第1、2、3周行肝脏MRI扫描观察移植细胞受体内定居增殖情况及受体肝内迁徙、定居情况.结果 菲立磁标记胚胎干细胞效率达到91.5%以上,体外有效引发MRI信号改变的最适移植细胞数量为1×105.肝脏MRI扫描,可观察到移植的标记细胞在受体肝内能定居并增殖,且与受体肝板有良好整合.结论 通过MRI活体示踪可以动态连续监测移植菲立磁标记胚胎干细胞体内的分布和增殖.

Abstract:Objective To investigate the ability of residence, proliferation and integratation of embryonic stem cells (ESCs) labeled with feridex after transplantation into liver-injured mouse liver.Methods The mice were divided into the liver-injured group and the control group. ESCs were transplanted via liver subcapsular injection. Magnetic resonance imaging (MRI) examination was done to track the transplanted cells after 1, 2 and 3 weeks. Results The feridex labeling efficiency of ESCs was above 91.5%. The number of optimal transplanted cells which induced MRI signal changes in vtiro was 1 × 105.MRI and pathological detection showed that the satisfactory ability of residence, proliferation and integratation of ESCs in injured liver. Conclusion ESCs labeled with feridex can settle down, proliferate and integrate into liver-injured mouse liver. Transplantation of cells labeled with feridex is a safe and efficient method for tracking ESCs in vivo.
