摘    要:目的:探讨多药抗药基因(mdr_1基因)表达与人肺腺癌多药抗药细胞系LC-3/CDDP抗药性产生的关系.方法:应用地高辛标记的mdr_1 cDNA探针,采用斑点杂交方法,检测该细胞系中的mdr_1mRNA含量,同时以免疫细胞化学染色方法,检测P-糖蛋白(P-gP)表达,并与亲代(LC-3)细胞比较.结果:LC-3/CDDP细胞存在mdr_1基因过表达.结论:mdr_1基因过表达是LC—3/CDDP细胞系产抗药性的机制之一.

Abstract:Purpose:Investigating whether mdr1 gene expression is related to resistance to anti-cancer drugs of a multidrug re-sisitant lung cancer cell line LC-3/CDDP. Methods:mRNA was detected by dot blot gybridization with Digoxingen labelled mdr1 cDNA probe, and P ?glycoprotein was also detected by immunocytochemistry with P-glycoprotein monoclonal antibody. Results: It was showed that mdr1 mRNA in LC-/CDDP cells was 39. 6 pg/10 cells, while in LC-3 cells was only 1. 92 pg/10 cells. P-gly-coprotein was highly expressed in LC-3/CDDP cells, but was negative in LC-3 cells. Conclusion: These results indicated that mdr1 gene is overexpressed in LC-3/CDDP cells, which suggests that mdr1 gene expression may be one mechanism for multidrug resistance of LC-3/CDDP cells line.
