基于网络药理学研究葛根芩连汤治疗溃疡性结肠炎的分子作用机制

摘    要:目的:通过网络药理学方法探讨葛根芩连汤治疗溃疡性结肠炎的具体分子机制.方法:通过TCMSP、BATMAN-TCM数据库对葛根芩连汤主要成分及潜在作用靶点进行收集.从DisGeNET数据库中寻找与溃疡性结肠炎相关的疾病基因与潜在靶点进行映射,得出该方治疗溃疡性结肠炎的最终作用靶标.将最终靶标输入STRING数据库得出"蛋白-蛋白"相互作用关系,并用Cytoscape软件进行可视化.输入DAVID数据库进行GO、KEGG PATHWAY富集,得出其可能作用的分子机制并构建"成分-靶标-信号通路"网络图.结果:通过网络药理学预测后发现,葛根芩连汤中有127个有效成分可用于治疗溃疡性结肠炎,其中涉及155个靶基因及包括TNF、NF-kB、HIF-1等在内的22个信号通路.结论:从网络药理学角度初步揭示了其治疗溃疡性结肠炎的分子机制,为后续的研究提供了基础.

Abstract:Objective:To probe the specific molecular mechanism of Gegen Qinlian Decoction in the treatment of ulcerative colitis by network pharmacological method.Methods:The main components and potential targets of Gegen Qinlian Decoction were collected through TCMSP and BATMAN-TCM databases.The mapping of disease genes and potential targets related to ulcerative colitis from the DisGeNET database was conducted to obtain the final target for the treatment of ulcerative colitis.The final target was input into the STRING database to derive the"protein-protein"interaction,which was visualized by Cytoscape software.The GO and KEGG PATHWAY enrichment was carried out in the DAVID database to obtain the possible molecular mechanisms and construct the"component-target-signalingpathway"networkmap.Results:According to prediction of the network pharmacology,there are 127 active ingredients in Gegen Qinlian Decoction can be used to treat ulcerative colitis,involving 155 target genes and 22 signaling pathways including TNF,NF-kB、HIF-1 and so on.Conclusion:the molecular mechanism of ulcerative colitis was preliminarily revealed from the perspective of network pharmacology,which provided the basis for the subsequent research.
