摘 要: | 目的:建立HPLC法测定广东紫珠干浸膏中咖啡酸的含量.方法:采用Agilent 5 TC-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.5%磷酸(18:82)为流动相,检测流速为1.0 mL·min-1,波长323 nm,柱温30℃.结果:咖啡酸的进样量在0.04~0.80μg(R2=0.9953)范围内呈良好线性关系;咖啡酸的平均回收率(n=6)为95.93%(RSD%=1.02);10个批次广东紫珠干浸膏样品中咖啡酸的含量测定结果分别为0.22~0.83 mg/g.结论:经方法学验证,本法可用于广东紫珠干浸膏中咖啡酸的含量测定.

Abstract: | Objective:To establish an HPLC method for the determination of caffeic acid in the dry extractum of Callicarpa kwangtungensis.Methods:The HPLC system consisted of an Agilent 5 TC-C18 column(4.6 mm×250 mm,5μm);The mobile phase consisted of acetonitrile-0.5%phosphoric acid(18∶82)and the flow rate was 1.0mL·min-1;The detection wavelength was set at 323 nm.The column temperature was set at 30℃.Results:The linear range was 0.04-0.80μg(R2=0.9953)for caffeic acid;The average recoveries(n=6)were 95.93%(RSD%=1.02);The contents in 10 samples were 0.22-0.83 mg/g for caffeic acid.Conclusion:According to the methodology validation,this method can be applied to the determination of caffeic acid in the dry extractum of Callicarpa kwangtungensis. |
