摘 要: | 目的建立测定贞芪扶正片中黄芪甲苷含量的高效液相色谱-蒸发光散射检测器法(HPLC-ELSD)。方法采用Kromasil C18(250mm×4.6mm,5μm)的色谱柱;以甲醇-水(80∶20)为流动相;流速:1ml/min;ELSD检测条件:检测器漂移管温度为80℃,氮气压力为50psi,柱温为30℃。结果黄芪甲苷在1.5225-15.225μg范围内与峰面积分别取自然对数后呈良好的线性关系(r=0.9994,n=6),平均回收率是99.98%(RSD=0.3%)。结论HPLC-ELSD法测定贞芪扶正片中黄芪甲苷含量简便、准确、可靠。

Abstract: | Objective To determine astragaloside Ⅳ in zhenqi fuzheng tablet by HPLC-ELSD. Methods HPLC-ELSD was used in the quantitative analysis of astragaloside Ⅳ in the sample. The separationwas performed on Kromasil C18 (250 mm × 4.6 mm, 5 μm) column with a mixture of methanol-water( 80: 20) as mobile phase and the temperature of column was 30℃. The flow rate was 1.0 ml/min. The temperature of drift tube for measurement was 80℃, and the flowrate of N2 was 50psi. Results The standard curve was linear in the range of 1. 522 5-15.22 5 μg for astragaloside Ⅳ (r =0. 999 4,n =6). The average recovery was 99. 98% and RSD was 0. 3 %. Conclusion The method is simple, reliable, and is a good method to determinate astragaloside Ⅳ. |
