山楂叶的化学成分

摘    要:目的分离、鉴定山楂叶乙醇提取物中的化学成分。方法山楂叶的体积分数为80%的乙醇水提取物经D101大孔树脂处理,用水和不同浓度的乙醇洗脱,体积分数为55%的乙醇水洗脱部分采用硅胶吸附柱色谱和制备高效液相色谱法进行分离并纯化,根据化合物的理化性质和光谱数据(IR、NMR)分析进行结构鉴定。结果分离得到5个化合物,分别鉴定为:2α,3β,19α三羟基熊果酸(2α,3β,19αtrihydroxyl ursolic acid,1)、熊果酸(ursolic acid,2)、牡荆素(vitexin,3)、2″O鼠李糖基牡荆素(2″Orhamnosyl vitexin,4)、山奈酚(kaempferol,5)。结论化合物2α,3β,19α三羟基熊果酸(2α,3β,19αtrihydroxyl ursolic acid,1)为首次从山楂属植物中分离得到,作者经过对化合物1的1H NMR1、3C NMR、HMQC谱和HMBC谱认真解析并对照已知文献,对文献中部分13CNMR谱信号的归属错误进行了纠正。

Abstract:Objective To study the chemical constituents of the extract from the leaves of Crataegus pinnatifida Bge.Methods The extract from the leaves of Crataegus pinnatifida Bge.were eluted by water and different concentration of alcohol on the D101 macroporous resin.The 55% alcohol-water portion was collected and chemical constituents were isolated by silica gel column chromatography and preparative HPLC.Physical-chemical characters and spectroscopic analysis were employed for the structural identification.Results Compound 1-5 were obtained and their structures were elucidated as 2α,3β,19α-trihydroxyl ursolic acid(1),ursolic acid(2),vitexin(3),2′-O-rhamnosyl vitexin(4),kaempferol(5).Conclusions 2α,3β,19α-trihydroxyl ursolic acid(1) is isolated from Crataegus pinnatifida Bge.for the first time.The part data of()~(13)C-NMR of compound 1 in the literature were corrected by the 2D-NMR spectroscopic methods.
