瘤内注射新城鸡瘟病毒的抗癌效应及其机理研究

摘    要:本文报道了瘤内注射新城鸡瘟病毒(NDV)对615系小鼠乳腺癌(Ca761)治疗作用,并初步探讨了其抗瘤机理。结果表明:单纯NDV瘤内注射有一定抑瘤作用,抑瘤率达35.3%;联合环磷酸胺化疗,可使其抑瘤率提高至74.1%,明显优于化疗对照组(P<001),而该病毒腹腔注射则无效。治疗后带瘤小鼠的瘤细胞电镜显示:NDV颗粒主要位于瘤细胞的胞浆内及胞膜上。Western印迹检测细胞膜上有该病毒相关抗原的表达,其分子量分别是:80kD、55kD、49kD、38kD、29kD。进一步以MTT比色分析法证实:治疗后小鼠脾细胞的CTL杀伤活性明显增强,且具有一定的肿瘤特异性,提示NDV瘤内注射可能通过肿瘤细胞“体内异种化”而增强机体的主动特异性抗癌免疫反应。

Abstract:In this paper,we reported the therapeutic effects of intratumorous injection of NDV(NDV I.T.) against a murine mammary cancer model Ca761 and preliminarily discussed the possible mechanisms for the anti-tumor effects.The results showed that to a certain degree tumor regression was obtained by NDV I.T.alone,and the tumor inhibition rate was 35.3%(P<0.05).Co-therapy with cyclophosphamide(CY)resulted in a significant increase of tumor inhibition rate up to 74.1%,superiorly to that of CY chemotherapy control group(P<0.01).However,no therapeutic effect could be noticed when NDV was injected intraperitoneally.TEM indicated that NDV was located on the surface membrane and in the cytoplasm of Ca761 cells taken from NDV I.T.group.Further,the NDV-associated antigen(VAA) was detected on the Ca761 cells through Western immunoblotting using rabbit anti NDV-Ab and the molecule weights were 80kD,55kD,49kD,38kD,29kD respectively.it was possible that the VAA expressed on Ca761 cells could xenogenize them in vivo.The cytotoxicity of spleen cells from NDV I.T.group of mice assayed by MTT method showed that the cytotoxicity against Ca761-86 enhanced markedly, and exhibited some degree of tumor specificity.Suggesting that specific immunotherapeutic effect could be induced by NDV I.T.
