MTT法检测纳米金粒子体外细胞毒性的研究

摘    要:目的研究球形纳米金粒子对人成纤维细胞的体外细胞毒性作用;方法应用柠檬酸钠还原法制备纳米金溶胶,将纳米金溶胶制备成不同浓度的含金培养液,采用MTT法体外检测其对人成纤维细胞的细胞毒效应。结果培养液中加入不同浓度纳米金溶胶(1.875 mg.L-13、.750 mg.L-17、.500 mg.L-11、5.000 mg.L-13、0.000 mg.L-1)各组细胞相对增殖率(RGR)分别为112.653%,111.293%,89.524%,72.109%,46.803%,加入15.000 mg.L-1、30.000 mg.L-1纳米金溶胶组,其RGR与空白对照组有显著差异,1.875 mg.L-1、3.750 mg.L-1组毒性评级为0级,7.500 mg.L-1、15.000 mg.L-1组毒性评级为1级,倒置显微镜下细胞形态正常,生长良好,30.000 mg.L-1组毒性评级为3级,镜下可见细胞明显的空泡变性;结论此种纳米金粒子的体外细胞毒性呈剂量依赖关系,低浓度纳米金粒子具有良好的生物相容性,30.000 mg.L-1纳米金粒子有中度细胞毒性。

Abstract:Objective To investigate the cytotoxicity of spherical gold nano-particles to human fibroblasts(HF) in vitro;Methods Human fibroblasts(HF) were cultured with medium contained different dose of spherical gold nano-particles(1.875 mg·L-1、3.750 mg·L-1、7.500 mg·L-1、15.000 mg·L-1、30.000 mg·L-1) for 24 hours,the cytotoxicity effect was tested by MTT method,morphology of HF cells was observed under the inverted microscope;Results The Relative growth rate(RGR)was 112.653%,111.293%,89.524%,72.109%,46.803% respectively in different dose of gold nano-particles groups,the the RGR in 15.000 mg·L-1,30.000 mg·L-1 group had significant difference compared with blank group(P0.05),the cytoxicity grade was 0 grade in 1.875 mg·L-1、3.75 mg·L-1 groups,1 grade in 7.5 mg·L-1、15 mg·L-1 groups and 3grade in 30.000 mg·L-1 groups.The HF had normal morphology and well growth under inverted microscope observation in low dose of gold nano-particles groups while obvious vacuolar degeneration in 30.000 mg·L-1 gold nano-particles group.Conclusion The cytotoxicity of the gold nano-particles had does-effect response,low dose of gold nano-particles has well biocompatibility with HF,and 30.000 mg·L-1 gold nano-particles has moderate cytotoxicity.
