星点设计-响应面法优化热水浸提北沙参多糖的工艺研究

摘    要:目的:采用星点设计-响应面分析法对热水浸提北沙参多糖的工艺进行优化。方法:热水浸提法提取北沙参多糖,采用苯酚-硫酸法测定多糖含量。以北沙参多糖提取率为指标,对浸提温度、时间、液料比、次数等影响多糖提取率的单因素进行考察。结合单因素考察结果进行3因素5水平的星点设计试验,以响应面法优化最佳工艺。结果:优选工艺为25倍量蒸馏水于94.9℃条件下浸提2次,每次120 min,提取实际值与理论值偏差为4.05%。结论:优化后的提取工艺稳定可行,预测性良好。

Optimization of Hot Water Extraction Method of Glehnia Littoralis polysaccharides by Central Composite Design and Response Surface Methodology

Abstract:Objective:To optimize extraction conditions of hot water technology of polysaccharides from Glehnia littoralis by central composite design and response surface methodology. Methods:Polysaccharides of Glehnia littoralis was extracted by hot water extraction method and the content of polysaccharides was determined by phenol sulfuric acid method. Polysaccharides extraction rate was used as the index,and single factors such as extraction temperature,extraction times,amount of water,times which could affect extract rate was studied. Based on single factor test results,3 factors and 5 levels central composite design was applied and response surface method was used to optimize the best process. Results:The optimal technology was as follows:in the 25 folds of water,the extraction temperature was at 94.9 ℃,and extract was performed for 2times,120 minutes per time. The difference between observed value and predicted value was 4.05% under this condition.Conclusion:The optimized extraction process is stable and feasible,and the prediction is good.
