新加香薷饮对湿热型流感小鼠的疗效机制研究

摘    要:目的:探讨新加香薷饮治疗湿热型流感小鼠的免疫学机制。方法 :将50只C57BL/6小鼠随机分为5组:正常组,湿热组,湿热+病毒组,湿热+病毒+利巴韦林组,湿热+病毒+新加香薷饮组。正常组在正常环境下饲养,湿热组在湿热环境下造模,其他3组在湿热环境下给予甲1型流感病毒(FM1株)滴鼻造模。造模成功后湿热+病毒+利巴韦林组、湿热+病毒+新加香薷饮组分别给予利巴韦林、新加香薷饮干预治疗,其他组给予生理盐水灌胃。解剖取材后进行小鼠左肺叶苏木精-伊红(HE)染色,右肺叶肺组织血凝素(HA)、Toll样受体7(TLR7)与髓样分化因子(MyD88)基因逆转录实时荧光定量聚合酶链反应(qRT-PCR)的检测。结果:成功构建湿热型流感小鼠模型,其TLR7、MyD88 mRNA表达量显著升高,与正常组相比,湿热型小鼠饮水量及摄食量下降,大便偏湿,肛门红肿。与湿热组相比,湿热+病毒组小鼠体质量下降、精神萎靡、扎堆、皮毛蓬松、挠鼻,肺组织充血肿大、肺泡壁增厚、大量淋巴细胞浸润,HA mRNA相对表达量比较亦有差异(P<0.05),并且TLR7、MyD88 mRNA表达显著上调(P<0.05)。与湿热+病毒组相比,湿热+病毒+利巴韦林组、湿热+病毒+新加香薷饮组体质量下降趋势较缓慢,活动量稍多,肺组织结构虽紊乱,但渗出液较少,无透明膜的形成,而TLR7、MyD88 mRNA与湿热+病毒组相比差异无统计学意义(P>0.05);与湿热+病毒+利巴韦林组相比,湿热+病毒+新加香薷饮组肺泡壁增生变厚程度较轻。结论:新加香薷饮可减轻病毒性肺炎的炎症反应,降低肺脏病理损伤,对湿热型流感小鼠有一定的治疗作用,然而其并未影响TLR7、MyD88基因的表达。

Mechanism Study on the Effects of Xinjia Xiangru Decoction on Mice with Damp-heat Type of Influenza

Abstract:Objective:To study the immunologic mechanism of Xinjia Xiangru Decoction in treating the mice with damp-heat type of influenza.Methods:Total 50 C57BL/6 mice were randomly divided into 5 groups:the normal group,damp-heat group,damp-heat and viral group,damp-heat,viral and ribavirin group,damp-heat,viral and Xinjia Xiangru decoction group.The mice in the normal group were raised in normal environment.The mice in the damp-heat group were raised in the damp-heat environment to establish models.The mice in the other 3 groups living in the damp-heat environment were injected FM1 virus through their nasal cavity to establish models.After models established,the mice in the damp-heat,viral and ribavirin group were treated with ribavirin and Xinjia Xiangru decoction was used to treat the mice in the damp-heat,viral and Xinjia Xiangru decoction group.The mice in the other groups were given saline lavage.After dissection,the left lung of mice was used for HE staining,while the other one was applied to the test of q RT-PCR for the HA,TLR7 and MyD88 genes.Results:The model mice with damp-heat type of influenza were established successfully and the expressions of TLR7 and MyD88 mRNA were rising obviously.Compared with the normal group,the mice with damp-heat type were in the condition of less water and food intake,wet stool,red and swelling anus.Compared with the damp-heat group,the mice from the damp-heat and viral group were loss of weight,apathetic,gathering together,fluffy fur and scratching their noses,while their lung tissue were hyperemia and swelling,their alveolar wall were thickening and infiltrated by large of lymphocyte.There was the significant difference between the damp-heat group and them on the expression of HA mRNA (P< 0.05).And the expressions of TLR7,MyD88 mRNA in the damp-heat and virus group increased obviously (P< 0.05).Compared with the damp-heat and viral group,the mice from the damp-heat,viral and ribavirin group and the damp-heat,viral and Xinjia Xiangru decoction group were slow in the trend of their weight,energetic slightly in their activity,there weren't the percolate or hyaline membrane although the structure of lung tissue was disordered.However,there were no statistical significance between these two groups and the damp-heat and viral group on the expressions of TLR7 and MyD88 (P> 0.05).Compared with the damp-heat,viral and ribavirin group,the alveolar wall of the damp-heat,viral and Xinjia Xiangru decoction group was thickening fewer.Conclusion:Xinjia Xiangru decoction can relieve the inflammatory response of viral pneumonia and reduce the pathological damage.The decoction makes a certain therapeutic effect on the mice with damp-heat type of influenza,but fsiled to affect the expressions of TLR7 and MyD88 genes.
