姜黄素对3T3-L1前脂肪细胞增殖分化的影响及机制研究

摘    要:目的:探讨姜黄素对3T3-L1前脂肪细胞增殖、分化的影响及其机制。方法:以XTT法检测3T3-Ll前脂肪细胞的增殖;油红O染色并通过比色定量分析检测3T3-L1前脂肪细胞分化过程中胞浆脂质的堆积,实时定量荧光PC(real-time-PCR)检测脂肪细胞增殖、分化相关基因内脂素、抵抗素、脂联素mRNA的表达。结果:姜黄素组A值显著高于空白对照组并随着剂量增加而增大,且具有显著性差异(P0.05);姜黄素作用细胞48 h时,促进或抑制细胞增殖的作用最强,40μmol/L以上浓度的姜黄素出现细胞毒性作用,大部分细胞成片脱落死亡;72 h时,15~35μmol/L姜黄素组细胞的生长率有所增加,而40μmol/L以下姜黄素处理组细胞生长率仍维持在较低水平;脂肪细胞油红O染色分光光度法结果显示2.5,5,10,15和20μmol/L姜黄素剂量组的细胞吸光度值与空白对照组相比有显著性差异(P0.05);姜黄素组、吡格列酮组的内脂素、抵抗素mRNA表达均较对照组显著降低,而脂联素较对照组明显升高,且具有显著性差异(P0.05)。结论:姜黄素能够促进前脂肪细胞分化,低浓度时促进其增殖,高浓度时则抑制其增殖,降低内脂素、抵抗素mRNA表达,促进脂联素mRNA的表达。

Abstract:Objective:To discuss the effects of curcumin on the proliferation and differentiation ot 3T3-L1 preadipocytes and mechanism. Methods:The proliferation of 3T3-L1 preadipocytes was detected by XTF method. Lipid droplets accumulated in cytoplasm of the differentiated preadipocytes were observed by using red O staining and quantified by colorimetry. The expressions of adiponectin,resistin and visfatin mRNAs were detected by real time polymerase chain reaction. Results:The result of curcumin group is higher than blank control group with significant difference(P〈0.05). The promoting or inhibiting effects of curcumin were the best on the 48 th hour,and there was cytotoxic effect at the concentration above 40 μmol/L with most ceils died in great amounts. The growth rate of cells in 15-35 μmol/L cureumin groups were in- creased at the 72 th hour,while that in curcumin groups at the concentration below 40 μmol/L were in lower levels.Results of oil red 0 staining showed that the absorbance in the 2.5,5,10,15 and 20 μmol/L cur- cumin groups were obviously increased than that of the blank control group(P〈0.05). Compared with control group,the expressions of visfatin and resistin mRNAs in curcumin group and pioglitazone group were signifi- cant decreased(P〈0.05),adiponectin was increased(P〈0.05). Conclusion:Curcumin can promote the prolifer- ation of 3T3-L1 preadipocytes,enhance the accumulation of lipid drops,and increase the terminal differentia- tion of preadipocytes,which may be associated with its effects improving the expressions of adiponectin,re- sistin and visfatin mRNAs.
