摘    要:目的 研究当归对脑缺血半暗带细胞凋亡的影响。方法 采用大鼠局灶性脑缺血再灌流模型,大脑中动脉阻塞2小时,再灌流48小时,用TUNEL法和免疫组织化学染色法分别检测使用当归后缺血半暗带凋亡细胞和凋亡相关基因Bcl-2、Bax蛋白表达。结果 与对照组比较,当归治疗组缺血半暗带的凋亡细胞显著减少(P<0.01);Bcl-2的表达显著增加(P<0.01),Bax的表达无显著变化(P>0.05)。结论 当归可能促进Bcl-2在脑缺血后的表达对半暗带的细胞凋亡产生抑制作用。

Abstract:Objective To investigate the effects of Angelica sinensis on apoptosis in ischemic penumbra following transient cerebral ischemia.Methods The middle cerebral arteries(MCA) of Wistar rats were occluded for 2 hours.The Angelica sinensis was administered intracavitary at 12g/kg.Apoptosis was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL).Bcl-2 and Bax expression was detected by immunohistochemistry method.Results In the Angelica sinensis group,the apoptotic cells were significantly decreased( P <0.01)and Bcl-2 protein expression was significantly increased( P <0.01) in ischemic penumbra than those mthe control group.Conclusion Angelica sinensis can prevent apoptosis in penumbra following cerebral ischemia due to the enhanced expression of Bcl-2.
