不同剂量地塞米松对SIRS大鼠心肌酶 CK-MB的影响

摘    要:目的观察不同剂量地塞米松对SIRS大鼠心肌酶CK-MB的影响。方法Wistar大鼠40只,体重160~200 g,雄性。随机分为5组:LPS注射后3 h组(3.0 h组)、LPS注射后6 h组(6.0 h组)、小剂量地塞米松治疗组(DXMⅠ组)、大剂量地塞米松治疗组(DXMⅡ组)、正常对照组,每组8只。实验组大鼠给予大肠杆菌内毒素(E.Coli O55B5,美国Sigma公司产品)20 mg/kg腹腔注射,制造SIRS大鼠模型,观察时间分别为3、6 h,DXM组在同样给予LPS后10 min给地塞米松,Ⅰ组给予10 mg/kg,Ⅱ组给予20 mg/kg腹腔注射,观察时间为3 h,结果与LPS 3.0 h组进行对比,对照组腹腔注射等量无菌生理盐水。观察SIRS大鼠各个时间心肌酶谱CK-MB的变化。结果LPS 3.0 h组、LPS 6.0 h组分别与对照组比较,结果有显著性差异(P均<0.01),尤以LPS 6.0 h组升高明显,说明SIRS大鼠心肌细胞受到严重损害,而且随着时间的推移,损害越来越重。与LPS 3.0 h组比较,DXMⅠ组CK-MB值下降明显(P<0.05),DXMⅡ组CK-MB值略有下降,但无显著性差异(P>0.05)。结论SIRS大鼠心肌受到损害,大剂量地塞米松对SIRS大鼠心肌损害的改善不明显,小剂量地塞米松对SIRS大鼠心肌损害的疗效优于大剂量地塞米松。

Abstract:Objective To explore the effects of different doses of dexamethason on systemic inflammatory response syndrome rat's cardiac CK-MB.Methods Forty Wistar rats were randomly divided into five groups:LPS(3.0 h) group,LPS(6.0 h) group,DXMⅠ group,DXMⅡ group,control group.The SIRS model of rats was made by lipopolysaccharide(LPS) injection,and DXMⅠ group received DXM(10 mg/kg),DXMⅡ group received DXM(20 mg/kg),intraperitoneally.The cardiac CK-MB of each group were observed and compared.Results Compared with the control group,the CK-MB of LPS(3.0 h) group,LPS(6.0 h) group were significantly increased(all P<0.01),Compared with LPS(3.0 h) group,the CK-MB of DXMⅠ group significantly decreased(P<0.05),but DXMⅡ group did not obviously decreased((P>0.05)).Conclusion The effects of(10 mg/kg) dose of dexamethason was better than(20 mg/kg) dose of dexamethason on improvement of SIRS rat's cardiac CK-MB.
