新疆地区三聚氰胺致尿结石动物模型研究

摘    要:目的:观察新疆地区高钙、高嘌呤饮食下管理限量内不同浓度三聚氰胺对大鼠草酸钙结石模型成石及肾脏损害的影响.方法:选270只SD大鼠,将大鼠随机分成27组,应用3×3析因实验设计,分别在高钙、高嘌呤饮食情况下,取三个剂量水平(0,0.5 mg/kg和1.0 mg/kg)三聚氰胺作为诱石剂,分别于8周、12周、16周测SD大鼠血常规及生化指标,并于16周采用麻醉法将SD大鼠处死,剖腹使用六六视觉体视显微镜系统观察SD大鼠的肾脏、输尿管和膀胱等器官的结石形成情况,分析比较高钙、高嘌呤饮食情况下,不同管理剂量三聚氰胺对SD大鼠肾功能及生化指标的影响及其对泌尿系形成解释的影响.结果:①在同一CnBn水平下,不同水平三聚氰胺对SD大鼠体质量的影响未见统计学差异;②A(三聚氰胺+碳酸钙)、B(三聚氰胺+腺嘌呤)和C(时间)三因素对SD大鼠血光抑素、血肌酐、血尿酸及血清钙的影响未显示存在交互作用(P>0.05).两因素间的交互作用中,B、C两因素对SD大鼠血光抑素、血肌酐、血尿酸及血清钙的影响存在交互作用(P<0.05);③16周时,不同剂量三聚氰胺+腺嘌呤饮食情况下,大鼠的血光抑素、血尿酸及血尿素氮的差异有统计学意义,出现早期肾功能损害;④不同因素Cx-AxBx各水平间,SD大鼠泌尿系形成结石情况比较显示其差异均无统计学意义(P>0.05).结论:限量范围内的三聚氰胺+腺嘌呤在一定时间的累计下可造成染毒16周的大鼠肾功能损害,但并未显示形成肾结石.

Abstract:Objective: Observe different limits concentrations of melamine in high calcium and high purine diet management to form calcium oxalate stones of rats model into stone and the effects of kidney damage in Xinjiang region. Method 270 SD rats were recruited in the study. Rats were randomly divided into 27 groups, useing 3 x3factorial experiment design, in the case of high calcium and high purine diet respectively, take three levels(0,0.5mg/kg and 1.0 mg/kg) of melamine as stone agent, SD rats blood routine and biochemical indexes were detected respectively in 8 weeks, 12 weeks and 16 weeks,and SD rats were killed using anesthesia at 16 weeks,and than observe SD rat kidney, ureter and bladder calculi formation, paunch useing 270th visual body depending on the microscope system. Finally analysis different doses of melamine management with high calcium and high purine diet,impacton SD ratrenalfunction and biochemicalindexand itsimpact on urinary tract formation interpretation. Results①under thesame CnBn level, the effect of different levels of melamine on the quality of the SD rat body has not seen statistically significan;②three factors including A(melamine + calcium carbonate), B(adenine +melamine) and C(time) on serum creatinine, uric acid and blood serum calcium of the lotus flower inhibition SD rats, did not show the influence of interaction(P〉0.05).The interaction between the two factors, B and C two factors on serum creatinine,blood uric acid, and the influence of serum calcium of the lotus flower inhibition SD rats exist interaction(P〈0.05);③the blood uric acid and blood urea nitrogen of lotus flower inhibition of rats in different doses of melamine + adenine diet were various,and the difference was statistically significant in early renal damage at 16 weeks; ④In the different factors between different levels of CxAxBxSD,the differences of rats urinary stone formation had no statistical significance(P〉0.05).
