甲胎蛋白抑制PTEN活性导致肝癌细胞耐受ATRA诱导的凋亡的研究

摘    要:目的研究分析甲胎蛋白对PTEN活性产生抑制引起肝癌细胞耐受ATRA发生凋亡。方法选取2012年11月一2013年11月期间本院诊治36例肝癌患者,将患者术后肝癌细胞制成标本。结果人体肝癌HepG2与Bel7402细胞均存在PTEN表达,经160μmol/L的ATRA处理24h后可加强这些细胞的PTEN的表达。Co—IP技术实验得知甲胎蛋白可与PTEN相结合,最终对ATRA产生影响。结论肝癌细胞中表达的甲胎蛋白可与PTEN相结合,最终发现AFP为肝癌细胞耐受ATRA的重要因子。

Abstract:Objective To study and analyze inhibition of PTEN activity by alpha fetal protein caus ed resistance to all trans retinoic acid of hepatoma cells. Methods Select in November 2012 to November 2013 in the hospital make a diagnosis and give treatment of 36 cases of liver cancer patients, the patients of postoperative liver cancer cell specimen. Results Human hvpatocellular carcinoma HepG2 and Bel 7402 cell expression PTEN, 24, 160 mu of mol/L ATRA treatment can strengthen the cells the expression of PTEN as a child. Co-IP technology experiment told that AFP can be combined with PTEN, ultimately affect ATRA. Conclusion The liver cancer cells express AFP can be combined with PTEN, eventually found the AFP for the most important factors of liver cancer cells resistant to ATRA.
