细菌纤维素和中草药提取液治疗矽肺的研究

摘    要:目的:在矽肺大鼠模型中评估使用细菌纤维素、细菌纤维素混合中草提取液治疗矽肺的效果。方法:在体重为180—220g的大鼠肺部注入50mg/只的二氧化硅粉尘给大鼠染尘;利用上述各种制剂对大鼠进行治疗,治疗四周后处死,通过观察组织病理学病变、检测脏体系数、全肺胶原含量、全肺干重、全肺游离二氧化硅含量以及检测肺灌洗液中细胞计数及分类来全面评估各种制剂的治疗效果。结果:各种制剂的治疗效果依次为:含细菌纤维素的中草药提取液治疗组〉细菌纤维素治疗组〉阳性对照组,但是,各检测指标数据与阳性对照组未发现有明显的统计学差异,说明两种制剂的治疗效果不是很明显(P〈0.05)。结论:细菌纤维素治疗组稍微低于灌注的二氧化硅粉尘量,这初步表明细菌纤维能有效促进二氧化硅粉尘中肺部组织中排出,这进一步验证了中草药红茶茵中能产生细菌纤维素的木醋杆菌具有生物排尘的机理。

Treatment of Silicosis in Rats by Spraying with Bacterial Cellulose and Herbal Extracts

Abstract:Objective:Evaluate the effects of spraying with bacterial cellulose and the mixture of bacterial cellulose and herbal extracts in a rat silicosis model. Methods:Silica dust (50 mg) was injected into the lungs of rats, which were then treated with one of the experimental treatments for a month. The rats were then killed and the effects of the treatments were evaluated by examining the extent and severity of the histopathological lesions in the animals' lungs, measuring their organ coefficients and lung collagen contents, determining the dry and wet weights of their lungs, and measuring the free silica content of the dried lungs. In addition, lavage was performed on whole lungs taken from selected rats, and the numbers and types of cells in the lavage fluid were counted. Results: The most effective treatment in terms of ability to reduce lung collagen content and minimize the formation of pulmonary histopathological lesions was bacterial cellulose combing with herbal extracts treatment, followed by bacterial cellulose treatment and positive control group, but there are significant statistical differences among two treatments in these parameters, which proved that there were no significant treatment effects (P 〈 0. 05). Conclusions:The free sihca levels in the lungs of animals of bacterial cellulose treatment group was slightly lower than those for bacterial cellulose and herbal extracts treatment group, which primarily suggested that bacterial cellulose could effectively promote silica dust discharge from lung tissue and further validated that the biological dust emission mechanism of xylinum in the herbal kombucha.
