| 摘 要： | 目的:观察黄荆子木脂素VBE-3诱导人卵巢癌CoC1细胞凋亡作用。方法:体外培养CoC1细胞和中国仓鼠卵巢CHO-K1细胞,碘化丙啶(PI)染色流式细胞术(FCM)分析细胞凋亡率,琼脂糖凝胶电泳观察细胞DNA梯形条带。结果:PI染色FCM分析发现VBE-3以浓度依赖方式诱导CoC1细胞凋亡(P<0.01),对CHO-K1细胞的作用弱。琼脂糖凝胶电泳观察结果表明10μmol/L的VBE-3和LY294002处理24小时,CoC1细胞呈现典型凋亡细胞的DNA梯形条带;而CHO-K1细胞未见DNA梯形条带。结论:VBE-3选择性诱导CoC1细胞凋亡作用与其阻断PI3K信号传导有关。

| Abstract: | Objective To investigate the induction of apoptosis in ovarian cancer CoC1 cells by VBE-3,a lignan from the acetoacetate extract of of Vitex negundo seed.Methods CoC1 cells and Chinese hamster ovary(CHO-K1) cells were cultured in vitro.The apoptosis of cells induced by VBE-3 was determined by DNA fragment assay and flow cytometry using PI staining.Results VBE-3 significantly induced apoptosis of CoC1 cells in a concentration-dependent,and had little effective on CHO-K1 cells.DNA fragment assay indicated tha... |
