栀子甘草豉汤、大黄黄连泻心汤及其合方对反流性食管炎模型大鼠氧化应激影响

摘    要:目的:探讨栀子甘草豉汤、大黄黄连泻心汤及其合方对混合性反流性食管炎大鼠食管黏膜及氧化应激的影响及作用机制。方法:将72只健康雄性SD大鼠随机分为假手术组、模型组、西药对照组、栀子甘草豉汤组、大黄黄连泻心汤组、合方组。假手术组行伪手术,其余各组采用食管十二指肠吻合术建立反流性食管炎模型。1周后,假手术组、模型组给予生理盐水灌胃,西药组大鼠灌胃莫沙必利(1.75mg/kg),各中药组分别灌胃栀子甘草豉汤(5.83g/kg)、大黄黄连泻心汤(3.62g/kg)、合方(4.78g/kg)。每周空腹称重1次。术后5周处死全部大鼠,取全段食管称重后计算食管系数,肉眼观察做大体评分,并作组织病理学观察;制备食管组织匀浆,测食管组织中超氧化物歧化酶活性及丙二醛含量。结果:栀子甘草豉汤组、合方组大鼠的体质量增加值较模型组显著,三方均可显著降低食管中MDA含量,提高食管SOD活力,降低食管系数,改善食管大体及病理组织评分,其中合方效果最显著。结论:栀子甘草豉汤、大黄黄连泻心汤及合方能提高食管组织的抗氧化能力,具有防治大鼠反流性食管炎的作用,合方效果最佳。

The Effects of Gardeniae, Sojae Praeparati and Glycyrrhizae Decoction, Pieplant and Goldthread Purging Heart Decoction and Integrated Decoction on Oxidative Stress of Reflux Esophagitis Rat

Abstract:Objective: To observe the effects of Gardeniae, Sojae Praeparati and Glycyrrhizae Decoction, Pieplant and Goldthread Purging Heart Decoction and the Integrated Decoction on reflux esophagitis and the mechanism of action of oxidative stress in rat' s esophagus mucosa. Method: 72 healthy Wistermale male rats were divided into six groups randomly, which were sham operated group, model group ,western medicine control group, group of Gardeniae, Sojae Praeparati and Glyeyrrhizae Decoction (GSGD), group of Pieplant and Goldthread Purging Heart Decoction(PGD) and group of Integrated Decoction. Sham operated group was done with pseud-operation, and the other groups were done into models of reflux esophagitis by esophagoduodenostomy. After 1 week,isotonic Na chloride was given to sham operated group and model group by intragastric administration, Mosapride was given to western medicine control group ( 1.75 mg/kg), corresponding Chinese crude drug was given to each Chinese drugs group which was GSGD (5.83 g/kg) ,PGD (3.62 g/kg) and ID (4. 78 g/kg). Weigh once on an empty stomach every week. After 5 days of operation execute all the rats, select esophago to weigh and calculate esophago quotient,observe macroscopically and score, and do histopathology observation. Prepare esophago tissue homogehate and determine Superoxide dismutase (SOD) activity and Malondialdehyde (MDA) content. Result:Increasing value of rat' s weight was more in group of GSGD and group of ID than in model group. The three decoctions decreased MDA content remarkably, increased SOD activity, cut down esophago quotient, and improved esophago body and score of pathological tissue. ID did best. Conclusion:GSGD, PGD and ID can improve anti-oxadation ability of esophago tissue, and prevent injury from reflux esophagitis. ID has the best effect.
