玻璃化法保存家兔股动脉条件的筛选

摘    要:目的 筛选玻璃化法保存家兔股动脉的最佳条件。方法  (1)运用正交设计和氯化三苯基四唑 (TTC)染色筛选出适合家兔股动脉玻璃化保存和低温冷冻保存的条件。 (2 )通过细胞培养了解玻璃化动脉的细胞活性。结果  (1)玻璃化动脉的活性为 82 .67% ;低温冷冻动脉为73 .2 9% (P <0 .0 1)。(2 )二者经培养所得到的细胞为平滑肌细胞。结论 玻璃化法保存动脉由于在降温和复温过程中没有冰晶形成 ,对动脉整体结构和活性影响小 ,因而玻璃化动脉具有较好的细胞活性。

Abstract:Objective To investigate the appropriate conditions for preservation of femoral arteries in rabbits by vitrification.Methods (1) By using the orthogonal design and TTC testing,the appropriate conditions for preservation of femoral arterial grafts in rabbits by vitrification and cryopreservation were screened out.(2) The vitreous arterial grafts done under the cell culture and the cellular viability was determined and compared between fresh arteries and frozen arteries.Results (1) The viability of vitreous arteries was 82.67%,which was significantly higher than 73.29% of frozen arteries ( P < 0.01). (2) The cells that cultivated from vitrified arteries and frozen arteries were smooth muscle cells.Conclusion These results provided the evidence that the technical procedures of vitrification is simple,and vitrification has less damage to cells and tissues because of ice free in the process of cryopreservation,so the cellular viability is also better than frozen arteries.
