表皮生长因子对黄体细胞孕酮生成的影响

摘    要:表皮生长因子(EGF)对卵巢黄体细胞功能的影响与机制仍不清楚.本实验用体外培养的大鼠卵巢黄体细胞观察了EGF对基础孕酮、cAMP、cGMP生成的影响.结果表明:1.EGF刺激基础孕酮生成,并呈剂量依赖性;2.EGF同时也刺激cAMP与cGMP生成的增加,提示EGF增加孕酮分泌的作用与EGF促进第二信使物质中cAMP、cGMP生成有关;3.酪氨酸蛋白激酶(TPK)抑制剂genistein可明显抑制EGF刺激的孕酮与cAMP生成.本研究提示EGF是经先激活TPK途径,之后再影响cAMP等第二信使系统,进而影响孕酮的生成.

Abstract:The mechanism and effect of epidermal growth factor (EGF) on function of corpus luteum are not clear. In this experiment the effect of EGF on production of progesterone, cAMP and cGMP in cultured luteal cells of rat ovary is studied. The results show:l. that EGF sitmulates basal production of progesterone in a dose -dependent pattern;2. that EGF increases the produc-tion of cAMP and cGMP significantly at the same time. Which suggests that the stimulation of EGF to progesterone is related to EGF - induced production of second messengers of cAMP and cGMP;and 3. that genistein,a kind of inhibitor of tyrosine protein kinase(TPK) .inhibits EGF -induced production of progesterone and cAMP markedly. The results indicate that EGF influences second messenger system,cAMP.and then production of progesterone via activation of TPK path-way.
