摘    要:为了探讨联合应用 Neurturin和神经生长因子对老年性痴呆模型鼠海马突触素的影响 ,本研究采用切断成年 SD大鼠左侧穹窿海马伞 ,建立隔 -海马胆碱能系统损害的痴呆模型 ,损伤 4周后 ,用免疫组化和图象分析技术等方法对大鼠海马突触素进行定量分析。结果显示 ,损伤对照组损伤侧海马 CA1 区多形层、辐射层、腔隙分子层和齿状回分子层突触素含量分别减少了42 .60 %、46.0 4%、5 7.3 6%和 5 5 .2 2 % ,损伤对照组与正常对照组之间有高度显著性差异 ( P<0 .0 1) ;NGF治疗组损伤侧海马CA1 区多形层、辐射层、腔隙分子层和齿状回分子层突触素含量分别只减少了 3 3 .64 %、3 8.2 5 %、3 8.42 %和 2 7.64 % ,NGF治疗组与损伤对照组之间有显著性差异 ( P<0 .0 5 ) ;联合治疗组损伤侧海马 CA1 区多形层、辐射层、腔隙分子层和齿状回分子层突触素含量分别只减少了 9.97%、8.0 5 %、14 .70 %和 4.2 8% ,联合治疗组与损伤对照组之间有高度显著性差异 ( P<0 .0 1)。结论 :联合应用 neurturin和 NGF促使老年性痴呆模型鼠海马突触素含量增多的效果较单用 NGF治疗好

Abstract:To study the effect of neurturin(NRTN) and nerve growth factor(NGF) on synaptophysin(SYH) in hippocampus of rats with fimbria-fornix(FF) transection, adult SD rats were subjected to left FF transection and received continuous infusion of NRTN/NGF or NGF into the lateral ventricle. After four weeks, the rat brains were processed for synaptophysin immunocytochemistry. In the present study, we used computer-assisted optical density (COD) measurements of synaptophysin immuno-staining. The results showed that the value of COD decreased by 42.60%, 46.04%, 57.36% and 55.22% respectively in the polymorphous, radiatum, lacunosum and the molecular layer of dentate gyrus(MDG) in lesion control group and decreased by 33.64%, 38.25%, 38.42% and 27.64% respectively in the polymorphous, radiatum, lacunosum and the molecular layer of dentate gyrus in NGF group, and there was a significant difference between NGF group and control group(P<0.05). But the value of COD decreased by 9.97%, 8.05%, 14.70% and 4.28% respectively in the polymorphous, radiatum, lacunosum and the molecular layer of dentate gyrus in NRTN/NGF group, and there was a greatly significant difference between lesion control group and NRTN/NGF group(P<0.01). The results suggest that the content of synaptophysin in hippocampus of rats with FF transection can increase more significantly after simultaneous infusion of NRTN and NGF compared with infusion of NGF.
