大鼠实验性脑室出血后LN及LNR-1在脑内的表达及意义

摘    要:目的观察大鼠脑室出血后层粘蛋白(LN)及其受体LNR-1在大鼠脑内的表达变化,探讨其与出血后慢性脑积水之间的关系。方法将大鼠随机分为3组:正常对照组、假手术对照组、实验组,将枸橼酸化的0.1ml自体静脉血注入实验组大鼠侧脑室内建立出血后慢性脑积水模型(假手术组注入0.1ml生理盐水),采用免疫组化方法观察出血后不同时间LN及LNR-1在脑内的表达变化。结果实验组30d出现脑积水为80%(4/5)、其余组别未出现慢性脑积水;实验组出血后LN在纵裂池、软脑膜、蛛网膜、硬膜、脑内毛细血管壁、蛛网膜下腔血管壁的表达在3d时增加,7d表达继续增加,14d表达最强,较对照组、假手术组显著增强(P〈0.01),30d仍强烈表达;实验组出血后LNR-1在脉络丛的立方上皮细胞胞浆及顶质膜、室管膜细胞、软脑膜细胞、蛛网膜细胞、硬膜、纵裂池、胶质细胞的表达增加。LNR-1在蛛网膜下腔的表达变化与LN基本一致。结论大鼠脑室出血后LN及LNR-1的表达上调可能参与蛛网膜纤维化的过程,从而在出血后慢性脑积水的形成中发挥重要作用。

Abstract:Objective To observe the expression of laminin and laminin receptor 1 in rat brains and to study its correlation with the posthaemorrhagic chronic hydrocephalus.Methods Intraventricular infusion of 0.1 ml citrated autologous blood was performed to establish the experimental models of posthaemorrhagic chronic hydrocephalus.The expression changes of laminin(LN) and laminin receptor 1(LNR-1) in rat brains after intraventricular hemorrhage were detected by immunohistochemical techniques.Results Thirty days after intraventricular hemorrhage,4 rats(80%) presented chronic hydrocephalus in the experimental group.After intraventricular hemorrhage,the increase of LN expression in experimental rat brains were found in slit cistern,pia mater,arachnoid,dura and vascular wall.On the 14 th day,the expression of LN reached the peak and demonstrated evident increase compared with the control group(P<0.01).The increase of LNR-1 expression in the experimental rat brains was found in cuboidal epithelium of choroids plexus,epithelium of pia mater,arachnoid,dura and vascular endothelial cell.On the 14 th day,the expression of LNR-1 reached the peak and had evident increase compared with the control group(P<0.01).Conclusion The increase of LN and LNR-1 expression after intraventricular hemorrhage in rat brains might be involved in the development of arachnoidal fibrosis and posthaemorrhagic chronic hydrocephalus.
