摘    要:目的母鼠孕期和哺乳期暴露NP,观察其对子代神经生长相关蛋白(GAP-43)的影响。方法将交配成功的31只孕鼠根据妊娠H期分层后随机分配到4组,即对照组,NP低、中、高剂量组(NP0、25、50、100mg/kg/day),单笼饲养,空腹灌胃,NP暴露时间为受孕第6天到出生后21d哺乳期结束(GD6-PND21),免疫组织化学法检测21d、60d皮质部GAP-43的变化,并分析上述变化与NP暴露之间的关系。结果高剂剂量组21d和60d龄的仔鼠皮质部GAP-43免疫反应阳性细胞平均数目、积分光密度低于同期对照组仔鼠(P〈0.05);GAP-43蛋白表达与NP暴露剂量有负相关关系,差异有统计学意义(r=-0.711、-0.568,P〈0.05)。结论NP孕期暴露影响子代大鼠脑组织GAP-43的表达,且剂量越高,GAP-43表达越低。

Abstract:Objective To observe the adverse effects of Nonylphenol (NP) on neuronal growth - associated pro- tein ( GAP - 43 ) expression in the brain cortex of pups exposed to NP during gestational and lactational periods. Methods 31 pregnant dams were randomly assigned to 4 groups. The treatment groups received gavage with NP at dose levels of 25 mg/kg/day (low dose), 50 mg/kg/day (middle dose), 100 mg/kg/day (high dose) and groundnut oil alone (vehicle control), respectively. The pregnant dams were housed individually and fed intra- gastrically. NP exposure time was limited from gestational day 6 to postnatal day 21. Alterations in GAP -43 in the brain cortex of rat pups at PND 21 and PND 60 was detected using immunohistochemistry, and the associa- tion between NP exposure and these alterations in GAP - 43 was analyzed. Results GAP - 43 - positive cells and integrated optical density (IOD) of pup cerebral cortex at PND 21 and 60 in high - dose NP treatment group were lower than the control offspring (P 〈 O. 05 ). A negative and significant relationship between NP exposure dose and GAP-43 protein expression was revealed ( r = -0. 711, -0. 568 ,P 〈0.05 ). Conclusion Exposure to NP during gestation might have an adverse effect on GAP -43 expression in the brain of pup rats, an inverse dose -response relationship was found between NP exposure and GAP- 43 expression.
