| 摘 要： | 目的探讨黄连、吴茱萸不同配比的水提物(CREF)对人胃癌细胞(SGC-7901)生长和凋亡的影响,找出两药的最佳配比。方法采用MTT比色法观察不同配比的CREF对SGC-7901细胞生长的影响。应用流式细胞仪检测细胞凋亡率和细胞线粒体膜电位的变化。结果 1 mg/ml不同配比的CREF作用SGC-7901细胞24 h和48 h后,对细胞生长有明显抑制作用(P<0.05),且细胞的抑制率与黄连的剂量呈正比。流式细胞仪PI单染亚二倍体峰分析结果显示,以1 mg/mL不同配比的CREF 1∶6、2∶5、3∶4、4∶3、5∶2、6∶1(黄连:吴茱萸)作用于SGC-7901细胞48 h后,细胞凋亡率分别为(8.50±1.59)%、(9.90±1.01)%、(17.15±1.68)%、(21.55±1.97)%、(34.10±1.06)%和(34.40±1.02)%,与对照组(1.69±1.91)%比较,差异均有统计学意义(P<0.01)。JC-1染色检测结果显示,不同配比的CREF作用于SGC-7901细胞6 h后,绿/红荧光比值显著降低,细胞线粒体膜电位下降。结论 CREF能明显抑制SGC-7901细胞生长并可诱导其凋亡,黄连:吴茱萸最佳配比为6∶1。
