摘    要:目的观察祛风通络方对阿霉素肾病大鼠Pododcin mRNA表达及足细胞足突形态的影响,探讨祛风通络方治疗肾病蛋白尿的分子生物学机制。方法 SD大鼠随机分为空白组和造模组,以阿霉素(6 mg/kg)尾静脉注射法造模,模型成功后将大鼠随机分为模型组、祛风组、激素组和苯那组,并给予相应干预。用RT-PCR法测定肾组织Podocin mRNA表达,透射电镜观察足细胞及足突形态变化。结果 (1)模型成功与空白组比较,造模组大鼠Podocin mRNA表达明显下调(P<0.01),足细胞足突融合甚至消失,足细胞有脱落、细胞数明显减少。(2)干预结束后与模型组比较,各干预组PodocinmRNA表达均上调(P<0.01);祛风组足细胞及足突形态改善明显。结论祛风通络方可能通过上调阿霉素肾病大鼠肾小球足细胞Podocin mRNA表达,改善足细胞及足突形态结构而达到防治蛋白尿的目的。
