摘    要:目的分析不同亚型丙型肝炎病毒(HCV)包膜糖蛋白E1和E2的变异性。方法从HCV基因数据库中获得不同亚型HCV包膜序列,并利用生物信息学方法进行核苷酸序列变异性、包膜糖蛋白亲水性性及糖基化位点分析。结果与其他亚型相比,1b亚型HCV包膜糖蛋白E1和E2区核苷酸的变异性最高,且E1区的变异性高于E2区;在E1区55,72位和102—112位氨基酸处1b亚型亲水性较大;1b亚型的包膜糖蛋白E1区N一糖基化位点数最多,且在60位氨基酸处单独有一糖基化位点。结论与E2区相比,E1区的高突变可能与1b型HCV致病性较强关系密切,且1b亚型E1蛋白的大量糖基化也可能存病毒的致病及角癌挑浼中有一审作用。

Abstract:Objective To analyze the variability of envelope glycoprotein (env) of hepatitis C virus.Methods HCV subtype 1a、1b,2a,2b and 3a nucleotide sequences, and amino-acid consensus sequence in the FASTA format were collected from the HCV Sequence Database (http://hcv. lanl. gov). Variation of the nucleotide sequence and the Hydrophilicity and glycosylation within envelope glycoprotein were analyzed. Results El and E2 region of HCV subtype 1b had the most variability, genetic variability of E1 was higher than that of E2 region. E1 region of HCV subtype 1b had higher hydrophilicity at position 55-72 and 102-112 of amino acid residues;E1 region of HCV subtype 1b had the most highest frequency glycosylation site and had a Nglycosylation site in position 60. Conclusions The high variability of El region and glycosylation may be associated with the pathogenicity of HCV subtype 1b.
