摘    要:目的:研究人脐血间质干细胞(UCB-MSCs)脑内移植后的分布和移行,为细胞移植治疗脑创伤奠定基础.方法:健康孕妇顺产脐血经分离、培养得到人UCB-MSCs,并用Hoechst33258标记后备用.自由落体撞击法制作的大鼠脑创伤模型,将已标记的人UCB-MSCs移植到大鼠的脑损伤区.一段时间后处死大鼠,取脑组织切片,直接在荧光显微镜下观察移植细胞的存活和迁移.结果:细胞移植到大鼠脑内能够长时间存活,移植细胞与宿主细胞有很好的相容性,移植后3周细胞发出蓝白色光,沿移植道呈条带状分布,细胞在条带中央较为密集,在条带周围细胞分布逐渐稀疏.移植后6周,损伤组织中可见荧光标记的细胞,细胞分布较3周时稀疏.结论:人UCBMSCs脑内移植后,能够成活并向脑损伤区迁移,与宿主脑组织整合在一起.

Abstract:Aim: To explore the migration and location of human umbilical cord blood mesenchymal stem cells (UCB-MSCs) after they were transplanted into adult brain tissue and to investigate the possibility of a new cell source for central nervous system transplantation.Methods:UCB-MSCs were derived from separated umbilical cord blood, then, cultured and labeled by Hoechst33258 and were stereotactically transplanted into adult rat striatum after experimental traumatic brain injury. After a period of time, the rats were sacrified and their brain tissue was sectioned to identify the transplanted cells under fluorescent microscope.Results: These cells lived well and showed good compatibility when they were grafted into adult brains. Three weeks after transplantation, the marked UCB-MSCs were giving out blue-white lights and the morphology was intact and cells were spreading along the two sides of needle canal. The nearer to canal,the denser to spread. At 6 weeks after transplantation, the distribution of cells near the canal was sparser than that of 3 weeks.Conclusion: UCB-MSCs could live and incorporate into host brains when they are transplanted into adult brain tissue.
