颞叶癫痫大鼠睡眠障碍的分析

摘    要:目的 分析癫痫大鼠的睡眠结构变化及不同睡眠时相特异性痫样放电的发放情况.方法 运用立体定位技术在大鼠右侧海马CA3区微量注射Kainic acid(KA)建立颞叶癫痫大鼠模型,在海马CA3区的同一位点微量注射等量的生理盐水作为对照组.定期视频实时监控大鼠行为,并在模型建立8周后脑电图机进行多导睡眠图(PSG)描记.结果 癫痫组大鼠在麻醉清醒后出现痫样发作,发作约在清醒6h后减弱,1周发作基本终止,但在4周后又逐渐出现自发性发作.8周后PSG描记显示,与对照组相比,癫痫大鼠深慢波睡眠时间由(11.15±0.93)s减少到(6.00±0.98)s(P<0.05),异相睡眠时间由(9.80±0.66)s减少到(2.69±1.00)s(P<0.05),癫痫大鼠睡眠时痫样放电比率比清醒时升高,增加(108.3±42.5)%.结论 癫痫发作能引起睡眠结构的改变,主要表现为觉醒次数增加,深慢波睡眠减少,异相睡眠减少,癫痫大鼠在睡眠时痫样放电几率明显增加.

The analysis of sleep disorders in temporal lobe epilepsy rats
Abstract:ObjectiveTo analyse the influence of epileptic seizures on sleep configuration and epileptic discharges in different stages of sleep in rats. MethodsWith the stereotaxic technology, kainic acid (KA) was microinjected into CA3 area of the right hippocampus to establish an epileptic animal model, while the same volume of normal saline was microinjected into the same site as control group. The rats were monitored in regular time after KA injected. The PSG was performed by an electroencephalograph on the 8th week after the model established. ResultsThe rats of epileptic model performed status epilepticus after recovery from anesthesia, which would last almost 6 hours and faded in one week. However, the onset of spontaneous seizures reappeared at the 4th week after KA administration. After 8 weeks, all of rats were performed with PSG. The slow wave sleep 1 (SWS1) was decreased from (11.15±0.93)s to (6.00±0.98)s (P<0.05) and the paradoxical sleep (PS) was decreased from (9.80±0.66)s to (2.69±1.00)s (P<0.05). The frequency of epileptic discharges in epileptic animal models was increased during sleep in (108.3±42.5)%. ConclusionEpileptic seizures play an important role in the configuration of sleep architecture. In epileptic animal models,the wake is enhanced,SWS1 and PS are declined,and the frequency of epileptic discharges is increased.
