摘    要:目的:优选色谱条件,用高效液相色谱法测定葛根中葛根素和大豆苷含量。方法:以乙腈(A)和二次蒸馏水(内含0.05%磷酸、0.05%三乙胺)(B)为流动相,流速0.5mLmin-1,检测波长为210nm,进行梯度洗脱。结果:葛根素、大豆苷与其它杂质的分离度较好,葛根素和大豆苷的标准回收率分别为95.7%和95.6%,相对标准偏差分别为2.1%和1.7%。结论:方法可用于葛根中葛根素和大豆苷的测定。

Study on Determination of Puerarin and daidzin inPuerariae Lobatae Radix by High Performance Liquid Chromatography Method

Abstract:Objective: High performance liquid chromatography method (HPLC) for the determination of puerarin in puerariae lobatae radix by optimizing chromatography conditions was studied. Method: The mobile phase consisted of acetonitrile (A) - water ( containing 0.05% H3PO4 and 0. 05% three ethylamine) . The flow rate was 0.5mL min - 1. Puerarin and daidzin was monitored at 210nm by gradient elution. Result: Separating degree of puerarin , daidzin and impurities was better. The recoveries of puerarin and daidzin were 95. 7% and 95.6%. The relative standard deviations were 2. 1% and 1.7%. Conclusion: The method was applied to the determination of puerarin and daidzin in puerariae lobatae radix with good result.
