口腔鳞状细胞癌主要组织相容性复合体Ⅰ类分子A和B的表达

目的研究主要组织相容性复合体Ⅰ类分子A和B(MICA、MICB)在口腔鳞状细胞癌(OSCC)患者外周血淋巴细胞和健康成人外周血淋巴细胞中的表达差异,以及MICA、MICB对OSCC细胞生物学的影响及其意义。方法提取OSCC患者和健康成人外周血淋巴细胞,从中提取RNA。采用聚合酶链反应(PCR)、反转录聚合酶链反应(RT-PCR)检测MICA、MICB在OSCC患者外周血淋巴细胞和健康成人外周血淋巴细胞的表达差异。结果 PCR显示,OSCC患者及健康成人外周血淋巴细胞均表达MICA mRNA、MICB mRNA,但RT-PCR结果显示目的基因在两者中表达差异较大,OSCC患者外周血淋巴细胞MICA mRNA表达水平相当于健康成人的(2.8795±0.1343)倍,MICBmRNA表达水平相当于健康成人的(2.3268±0.1367)倍,均显著高于健康成人(P<0.01)。结论 OSCC外周血淋巴细胞MICA mRNA、MICB mRNA表达显著高于其在健康成人外周血淋巴细胞中表达。

Expression of MICA/B in oral squamous cell carcinoma

Objective To investigate the differences of MICA and MICB expression in peripheral blood lymphocytes(PBL) of patients with oral squamous cell carcinoma(OSCC) and healthy adults.To study the biological function and significance of MICA and MICB expression in OSCC.Methods Extrat PBL from patients with OSCC and healthy adults.MICA and MICB mRNA expression in PBL of patients with OSCC and healthy adults were detected by PCR/RT-PCR.Results In PBL,PCR results showed MICA/B mRNA were both expressed in patients with OSCC and healthy adults.RT-PCR results showed that purpose gene had different expression between the two groups.In PBL,the MICA mRNA expression of patients with OSCC was significantly higher than that in healthy adults,the MICB mRNA expression of patients with OSCC was significantly higher than that in healthy adults.Conclusion In PBL of patients with OSCC,MICA/B mRNA expression were significantly higher than that in healthy adults.
