狼疮样BXSB小鼠脾细胞CD134/CD134L的表达及治疗影响

摘    要:为了探讨CD134/CD134L共刺激信号在SLE发病机制中的可能作用及治疗的影响。采用狼疮样BXSB小鼠模型,随机分为三组:狼疮方治疗组、强的松治疗组、未治疗组,每组6只,疗程10周。另设与BXSB小鼠同基因的正常C57BL/6小鼠6只为正常对照组。分别采集上述各组小鼠外周血和脾组织进行检测。结果:(1)与正常对照组比较,未治疗组BXSB小鼠的血清IgG和抗dsDNA抗体水平和脾组织CD4+CD134+、CD8+CD134+、CD19+CD134L+、CD23+CD134L+的百分比及CD134、CD134L的mRNA拷贝数都显著增高(P<0.05或P<0.01)。且血IgG和抗dsDNA抗体水平与脾细胞CD19+CD134L、CD23+CD134L的水平增加呈正相关;(2)狼疮方治疗组或强的松治疗组的血清IgG、抗dsDNA抗体水平和脾组织CD4+CD134+、CD8+CD134+、CD19+CD134L+、CD23+CD134L+的百分比及CD134、CD134L的mRNA拷贝数都显著低于未治疗组(P<0.05或P<0.01),而与正常对照组无显著差异(P均>0.05)。狼疮样BXSB小鼠存在CD134/CD134L的异常表达。狼疮方可减轻狼疮样小鼠高丙种球蛋白血症,减少体内自身抗体产生,对CD134/CD134L的下调作用与强的松相当。

Abstract:To investigate the effects of treatment with Chinese medicine recipe for lupus on the expression and protein synthesis of CD134/CD134L in splenic cells of lupus-prone BXSB mice,80 male lupus-prone mice used were divided into 3 groups;(i.e.) untreated group,treated group with lupus recipe and the predisone-treated group,and were killed 10 weeks later.Six normal syngeneic C57BL/6 mice were to be used for control.RT-FQ-PCR was used for the detection of the mRNA expressions of CD134 and CD134L in the splenic cells of mice,and the percentages of the CD4~ CD134~ ,CD8~ CD134~ ,CD19~ CD134L~ and CD23~ CD134~ splenic cells were measured by flow cytometry analysis.Meanwhile,the serum levels of total IgG and anti-dsDNA were detected by ELISA.The experimental results showed that:(1) The serum levels of total IgG and anti-dsDNA antibody in untreated group were higher than those in the other groups,and there was no significant differeces among treated group with lupus recipe,predisone-treated group and control group.(2) The percentages of mRNA expressions on CD4~ CD134~ ,CD8~ CD134~ ,CD19~ CD134L~ and CD23~ CD134L~ splenic cells from untreated group of mice were obviously higher than those from the control group.(3) There was a positive correlation between the serum levels of total IgG and anti-dsDNA antibody in the untreated group with positve cell percentages of CD19~ CD134L~ and CD23~ CD134L~ cells.(4) In the lupus recipe-treated or predisone-treated groups of mice,the positive cell percentages as well as the mRNA expressions of the CD4~ CD134~ ,CD8~ CD134~ ,CD19~ CD134L~ and CD23~ CD134L~ cells were significantly reduced in comparison with the control group,while the difference between the treated and the normal group was not significant.It is concluded that abnormal expression of CD134/CD134L exists in the lupus-prone BXSB mice,and the treatment of the Chinese medicine recipe for lupus can reduce the production of IgG and the dsDNA antibody in the lupus-prone mice and the production of autoimmune antibodies in vivo.Its down-regulatory effect on CD134/CD134L is equivalent to that of predisone.
