MAGE-1、MAGE-3基因在胃癌和胃活检组织中的表达及意义

摘    要:目的 检测黑色素瘤抗原基因-1、 -3(MAGE-1和MAGE-3)在胃癌中的表达, 探索其在胃癌免疫治疗中的应用价值.方法 采用RT-PCR法, 对36例胃癌患者的癌组织和相应的癌旁"正常"黏膜, 以及37例慢性胃炎患者的胃镜活检组织中, MAGE-1和MAGE-3基因的表达进行研究, 并对RT-PCR扩增产物中的目的基因片段进行DNA测序验证.结果 36例胃癌组织中, MAGE-1基因的表达阳性率为72.22%(26/36), MAGE-3基因的表达阳性率为69.44%(25/36).MAGE-1、 MAGE-3的表达均为阳性者为52.78%(19/36).癌旁"正常"黏膜中, MAGE-1、 MAGE-3基因的表达阳性率分别为47.22%(17/36)和44.44%(16/36); MAGE-1、 MAGE-3基因表达双阳性者为30.55%(11/36).在37例胃慢性炎症患者中, MAGE-1、 MAGE-3基因的表达阳性率, 分别为29.73%(11/37)和27.03%(10/37); MAGE-1、 MAGE-3基因的表达双阳性者为8.1%(3/37).胃癌组织中, MAGE-1和MAGE-3基因表达阳性率均高于癌旁和胃慢性炎症患者; 在肿瘤组织中的表达阳性率与肿瘤浸润的深度和分化程度有关(P<0.05); 而与临床病理分期及淋巴结转移无关(P>0.05).结论 基于MAGE-1和MAGE-3基因在胃癌中的高表达率, 可利用这两种蛋白作为靶分子进行免疫治疗, 同时也可作为一种临床有用的随访指标.

Abstract:AIM: To detect the expression of melanoma antigen 1 and 3 genes (MAGE 1 and MAGE 3) and explore its significance in immunotherapatic application. METHODS: The expression of MAGE 1 and MAGE 3 in gastric cancer, para tumorous normal mucosa, and gastric biopsy tissues were detected by RT PCR. The target genes in RT PCR product were verified by means of DNA sequencing. RESULTS: MAGE 1 expression was positive in 26 out of 36 (72.22%) cases of gastric cancer, MAGE 3 positive in 25 out of 36(69.44%), and both MAGE 1 and MAGE 3 positive in 19 out of 36 (52.78%). Positive expression rates of MAGE 1, MAGE 3 and both MAGE 1 and MAGE 3 were 47.22%, 44.44% and 30.55%, respectively in para tumorous normal mucosa, while 29.73%, 27.03% and 8.1% in gastric biopsy tissues The positive rates of MAGE 1 and MAGE 3 gene expressions in tumor tissues was significant higher than those its parallelism mucosae and biopsy tissues from chronic gastritis. The expressions of MAGE 1 and MAGE 3 genes in tumor tissues were relevant to intruded depth and the differentiated degree of tumor cells ( P <0.05). CONCLUSION: MAGE 1 and MAGE 3 protein may be used as targets molecules of immunotherapy for gastric cancer and indexes for follow up study of chronic gastritis as well.
