Abstract: | Our previous studies showed that geniposide (GP) inhibits aflatoxin B1 (AFB1)-induced hepatoxicity in rats, and that DNA repair synthesis in vitro and penta-acetyl geniposide ((Ac)5-GP) inhibits AFB1-induced genotoxicity in C3H10T1/2 cells. One possible mechanism for GP is the enhancement of the enzyme activity of glutathione S-transferase (GST) and GSH-peroxidase (GSH-Px) in AFB1-treated hepatocyte culture. But the mechanism is unknown for (Ac)5-GP. The present study demonstrated that (Ac)5-GP was more potent in inhibiting AFB1-induced unscheduled DNA synthesis in rat primary hepatocyte. The action of GP and (Ac)S-GP may be related to their ability to induce the activity of phase II enzymes. |
