| 摘 要： | ＰＣＲ指纹图技术选用通用型引物扩增ＨＬＡ－ＤＲＢ基因第二外显子，其产物在ＰＡＧＥ电泳中显示特定的“卫星”条带。可用于个体间作ＨＬＡ－ＤＲ的基因水平交叉配型，确定供受体间ＤＲＢ基因的相容性。采用ＨＬＡ纯合的Ｂ淋巴细胞系，分析单倍型上ＤＲＢＩ、ＤＲＢ３等基因异同时指纹图格局的变化，并通过１５对细胞间组合和比较，证实经ＤＮＡ交叉配合后的指纹图格局能十分敏感地反映个体间ＤＲ基因间的差异。在此基础上进一步对１７对随机个体作ＰＣＲ指纹图及交叉配型分析，表明这一技术在判别供受体间ＤＲ基因匹配性上十分有效，并有简单、快速、准确等优点。

| Abstract: | PCR-fingerprinting,which is based on PCR amplification of HLA-DRB genes exon 2 with universal primers,and comparing the electrophoresis patterns after PAGE,can determine the compatibility of a set of HLA-DRB genes simultaneously.The study shows in HLA either homozygous BLCLs or heterozygous individuals,only when the specificities of the haplotypes are identical,the fingerprints will be same,if the specificities are different,there will be different fingerprints and crossmatched fingerprints will creat new patterns. |
