家蝇幼虫抗茵肽粗提物对人肝癌HepG2细胞凋亡、钙离子浓度及线粒体膜电位变化的影响

摘    要:本文从肿瘤细胞凋亡及作用机制的角度出发初步探讨了家蝇幼虫抗菌肽粗提物对人肝癌HepG2细胞的影响.采用Hoechst 33258荧光染色法在荧光显微镜下观察家蝇抗菌肽对HepG2细胞形态学的影响.并通过流式细胞术分别观察家蝇抗菌肽粗提物对HepG2细胞凋亡、钙离子浓度及线粒体膜电位变化的影响.Hoechst 33258荧光染色法结果显示家蝇抗菌肽粗提物作用于HepG2细胞48h后能够引起细胞核发生核凝聚、染色质趋边化、核碎裂等凋亡形态学的改变;流式细胞仪检测结果显示家蝇抗菌肽粗提物能够诱导HepG2细胞发生凋亡、线粒体膜电位下降、细胞内钙离子浓度增加的现象.因此,我们推测家蝇抗菌肽可能通过增加HepG2细胞内钙离子,降低线粒体膜电位,进而诱导细胞凋亡的发生.

Abstract:In this paper,we investigate the effect of antimicrobial peptides extracts on HepG2 cells from the perspective of tumor cell apoptosis and its mechanism.Hoechst 33258 staining were used to observe the cell nuclear morphology of HepG2 after treatment with antimicrobial peptide from Musca domestica larva.Flow cytometry were used to analysis the effect of antimicrobial peptide on cell apoptosis,mitochondrial membrane potential and intracel|ular calcium concentration of HepG2 cells.HepG2 cells treated with Musca domestica antimicrobial peptides had a typical morphological characteristic apoptosis,such as nuclear cohesion,chromatin chemotaxis and nuclear fragmentation.HepG2 cells apoptosis rate were increased with decreased mitochondria membrane potential and increased calciumion concentration under the pressure of antimicrobial peptides from domestic flies.
