重组人促红细胞生成素后处理对兔缺血/再灌注骨骼肌细胞凋亡及超微结构的影响

摘    要:目的探讨重组人促红细胞生成素(rhEPO)后处理对家兔缺血/再灌注(I/R)骨骼肌细胞凋亡及超微结构的影响。方法将24只成年家兔随机分为对照绀、I/R组和rhEPO后处理组(5000IU/kg),采用25%的乌拉坦4mL/kg耳缘静脉注射麻醉,在I/R组、rhEPO后处理组建立兔左后肢腓肠肌I/R实验模型,4h后松开动脉夹,恢复血流灌注,12h后切取腓肠肌标本,采用原位末端标记法检测3组细胞凋亡指数,对I/R早期骨骼肌病理标本进行光镜及电镜观察。结果rhEPO后处理组细胞凋亡较I/R组明显减少,差异有统计学意义(P〈0.05);电镜及光镜下腓肠肌组织病理改变显示,rhEPO后处理组病变程度明显轻于I/R组。结论rhEPO后处理可通过减少细胞凋亡,改善骨骼肌I/R损伤。
