PUVA治疗银屑病Caspase3、bcl-2和PCNA在皮损组织中的表达

目的探讨银屑病经光化学疗法（PUVA）治疗后细胞增殖和凋亡相关指标表达的趋势。方法应用免疫组化技术检测30例寻常型银屑病经PUVA治疗前后及10例正常皮肤中PCNA、Bcl-2、Caspase3的表达。结果在正常表皮中Bcl-2和PCNA在基底层阳性表达，但在寻常型银屑病皮损中Bcl-2表达明显低于正常皮肤组，皮损经PUVA治疗后的表达恢复，有显著性统计学意义（P〈0．01）；PCNA在银屑病皮损为角质形成细胞广泛阳性表达，皮损经PUVA治疗后表达恢复，治疗前后有显著性统计学意义（P〈0．01）；Caspase3在寻常型银屑病皮损与正常皮肤的表达无显著性统计学意义（P〉0．05），皮损处与经PUVA治疗后皮肤的表达相比有显著性统计学意义（P〈0．01）。结论银屑病经PUvA后，Caspase3表达增加，PCNA、Bcl-2、表达恢复正常。

Expression of Caspase-3,Bcl-2,PCNA in Psoriatic Lesion Following the PUVA Therapy

Objective To explore expression of psoriatic apoptosis-related molecules,Bcl-2 Caspase-3 and proliferating cell nuclear antigen/cyclin(PCNA) in series of psoriatic lesion and psoriasis lesion following PUVA therapy and to evaluate the role of functional subsets of psoriasis cell proliferation and apoptosis.Methods Caspase-3,Bcl-2,PCNA were examined with immunohistochemical SP method in 30 cases of psoriatic lesion and psoriasis lesion following PUVA therapy and 10 cases of normal skin.Results Bcl-2 and PCNA in the normal skin just expressed in stratum basal and hardly expressed in stratum granulosum,demonstrating decreased Bcl-2 expression in psorisis compared with normal skin(P<0.01)and skin following PUVA therapy(P<0.01).Epidermal keratinocytes of psoriasis vulgaris was extensive expressed PCNA;PCNA in the psoriasis lesion following PUVA therapy was significantly decreased.PCNA expression in psoriasis was significantal following PUVA therapy than in psoriatic lesion(P<0.01).The results showed that Caspase-3 expression in psoriasis was no significant difference compared with normal skin(P>0.05),Caspase-3 expression in psoriatic lesion was significantly higher than skin following PUVA therapy(P<0.01).Conclusion The results showed that PCNA expression increased and Bcl-2 expression decreased when some factors induced to psorisis and lead to cell hyperproliferation.Caspase-3 expression was increased.PCNA and Bcl-2 expression were resumed in psoriasis by PUVA treatment.These results indicated that PUVA treatment induced apoptosis and resumed cell proliferation and cell death leading to structural homeostasis.
