不同时机针刺对卵巢早衰模型大鼠中枢及外周激素水平的影响

摘    要:目的探讨不同时机针刺对卵巢早衰模型大鼠中枢及外周激素水平的影响。方法50只清洁级性成熟未交配雌性SD大鼠,随机分为模型组(10只)、预针刺组(10只)、针刺组(10只)、佐剂组(5只)、空白组(5只)。模型组、针刺组及预针刺组连续15 d腹腔注射去氧乙烯基环己烯(4-vinylcyclohexene diepoxide,VCD)160 mg/kg建立卵巢早衰动物模型;佐剂组连续15 d注射同等剂量佐剂二甲基亚砜(dimethyl sulfoxide,DMSO);预针刺组在开始造模时即开始针刺,造模成功后继续治疗4周;针刺组于造模成功后开始针刺治疗,连续治疗4周;空白组不干预。治疗结束后,采用酶联免疫吸附测定(enzyme-linked immunosorbent assay,ELISA)试剂盒检测各组下丘脑促性腺激素释放激素(gonadotropin release hormone,GnRH)、垂体促卵泡生成素(follicle stimulatin hormone,FSH)、垂体促黄体生成素(luteotropic hormone,LH),卵巢中促卵泡生成素受体(follicle stimulatin hormone receptor,FSHr)、促黄体生成素受体(luteotropic hormone receptor,LHr)、雌激素受体(estradiol receptor,Er)、雌二醇(estradiol,E2),血清中GnRH、FSH、LH和E2水平。结果VCD腹腔注射可以造成大鼠卵巢早衰的动物模型,成功率为83.3%。与空白组比较,模型组大鼠下丘脑GnRH含量升高(P<0.05),垂体FSH和LH含量均升高(P<0.05);血清中GnRH、FSH、LH含量均升高(P<0.05),E2含量降低(P<0.05);卵巢组织中FSHr、LHr含量均升高(P<0.05),Er和E2含量均降低(P<0.05)。与模型组比较,预针刺组和针刺组大鼠下丘脑GnRH含量降低(P<0.05),垂体FSH含量降低(P<0.05);预针刺组和针刺组大鼠血清中GnRH、FSH含量均降低(P<0.05);预针刺组和针刺组大鼠卵巢组织中FSHr、LHr含量均降低(P<0.05),E2含量升高(P<0.05)。预针刺组和针刺组各项指标组间比较差异均无统计学意义(P>0.05)。结论针刺对POF模型大鼠中枢及外周激素水平均可产生一定的调节作用,不同时机针刺组间比较无差异。

Effect of Acupuncture at Different Times on Central and Peripheral Hormone Levels in a Rat Model of Premature Ovarian Failure

Abstract:Objective To investigate the effect of acupuncture at different times on central and peripheral hormone levels in a rat model of premature ovarian failure.Method Fifty clean-grade sexually mature non-mating female SD rats were randomized to model(10 rats),pre-acupuncture(10 rats),acupuncture(10 rats),adjuvant(5 rats)and blank(5 rats)groups.An animal model of premature ovarian failure was made by intraperitoneal injection of 4-vinylcyclohexene diepoxide(VCD)160 mg/kg for 15 consecutive days in the model,acupuncture and pre-acupuncture groups.The adjuvant group was injected with the same dose of Dimethyl sulfoxide(DMSO)for 15 consecutive days.The pre-acupuncture group received acupuncture before model making and four consecutive weeks of treatment after successful model making.The acupuncture group received four consecutive weeks of acupuncture treatment after successful model making.The blank group was not given intervention.Hypothalamic gonadotropin-releasing hormone(GnRH),pituitary follicle stimulating hormone(FSH)and luteotropic hormone(LH),ovarian follicle stimulating hormone receptor(FSHr),luteotropic hormone receptor(LHr),estradiol receptor(Er)and estradiol(E2),and serum GnRH,FSH,LH and E2 levels were measured by enzyme-linked immunosorbent assay(ELISA)in every group after the completion of treatment.Result Intraperitoneal injection of VCD could make a rat model of premature ovarian failure with a successful rate of 83.3%.Compared with the blank group,hypothalamic GnRH content increased(P<0.05),pituitary FSH and LH contents increased(P<0.05),serum GnRH,FSH and LH contents increased(P<0.05),serum E2 content decreased(P<0.05),ovarian LHr and Er contents increased(P<0.05)and ovarian E2 and Er contents decreased(P<0.05)in the model group of rats.Compared with the model group,hypothalamic GnRH content decreased(P<0.05),pituitary FSH content decreased(P<0.05),serum GnRH and FSH contents decreased(P<0.05),ovarian LHr and Er contents decreased(P<0.05)and ovarian E2 content increased(P<0.05)in the pre-acupuncture and acupuncture groups of rats.There were no statistically significant differences in various indicators between the pre-acupuncture and acupuncture groups(P>0.05).Conclusion Acupuncture can produce a certain regulating effect on central and peripheral hormone levels in a rat model of POF.There is no difference between the groups of acupuncture at different times.
