次声对大鼠焦虑抑郁情绪的影响

摘    要:目的:探讨不同暴露时程16 Hz、115 dB次声对大鼠焦虑抑郁情绪的影响。方法:健康成年雄性SD大鼠随机分为空白对照组(control)、假次声暴露组(sham exposure)和次声暴露组(infrasound exposure),空白对照组动物正常饲养于清洁级动物房,次声暴露组动物每天同一时间段置于次声实验室进行16 Hz、115 dB次声连续暴露(时程分别为7、14、21 d,2 h/d),假次声暴露组动物每天置于同一次声实验室但无次声暴露。大鼠焦虑抑郁情绪变化采用开放旷场和高架十字迷宫行为实验进行评价。结果:两组行为检测均显示,空白对照组与假暴露7、14、21 d组各行为指标均未发现统计学差异,表明次声实验室环境对大鼠行为学表现不会造成明显影响。开放旷场行为实验结果显示,次声连续暴露7 d组动物中央区运动距离显著低于假暴露7 d组(P <0.01),其他行为学实验指标未发现统计学差异;次声连续暴露14 d组动物中央区运动距离、中央区停留时间、中央区进入次数、总运动距离均显著低于假暴露14 d组,角落区停留时间、总静止时间则显著高于假暴露14 d组(P <0.05...

Abstract:Objective: To investigate the effects of infrasound exposure of 16 Hz and 115 dB with different duration on anxiety and depression in SD rats.Methods: Healthy adult male SD rats were randomly divided into control group,sham infrasound exposure group and infrasound exposure group.The animals in control group were normally feeded in clean rooms.Animals of infrasound exposure group were placed in the infrasound laboratory to accept continuous infrasound exposure of 16 Hz and 115 dB for 7,14,21 d(2 h/d) at the same time of each day.And the animals in the sham infrasound exposure group were placed in the same laboratory environment but without infrasound exposure.The changes of anxiety and depression in rats were evaluated by open field test or elevated plus-maze.Results: There was no statistical difference for each behavioral detection index between control group and sham infrasound exposure group for 7,14 d and 21 d,indicating no effect from the infrasound laboratory environment.The results of the open field test showed that,the movement distance of central field in the infrasound exposure group for 7 d was significantly less than that in the sham exposure group for 7 d(P < 0.01);whereas,no significant differences were found among other detection indexes between these two groups.On the other side,the movement distance of central field,the duration in central field,entry times of central field and total distance in infrasound exposure group for 14 d were significantly less than those in the sham exposure group for 14 d;the duration and total immobility time in corner field in infrasound exposure group for 14 d were significantly higher than those in the sham exposure group for 14 d(P < 0.05).The results of the elevated plus-maze behavioral experiment showed that the duration in close arm of infrasound exposure group for 7 d was higher than that in the sham exposure group for 7 d(P < 0.01);whereas,no significant differences were found among other detection indexes between these two groups.On the other side,the duration in close arm and the distance in close arm of infrasound exposure group for 14 d was higher than those in sham exposure group for 14 d;the entry times of open arm,percentage of the entries of open arm,the duration in open arm and percentage of the duration in open arm were all significantly less than those in the sham exposure group for 14 d(P < 0.05).No statistical difference was found in behavioral detection indexes between groups for 21 d.Conclusion: Infrasound exposure of 16 Hz and 115 dB for 7 or 14 d could induce anxiety and depression in rats,and the effect of exposure on 14 d was the most obvious.With the extension of exposure duration,anxiety and depression were alleviated.
