DNS法测定玉米须保健饮料中总多糖含量

目的考察供试品溶液制备的最佳条件,测定玉米须保健饮料中多糖含量。方法采用正交实验确定供试品溶液制备的最佳条件,二硝基水杨酸法显色,以葡萄糖为对照品,测定样品中总多糖的含量。结果葡萄糖质量浓度在8~56μg.mL-1范围内与吸收度呈良好的线性关系;平均回收率为101.83%,RSD=1.84%;测得的总多糖平均含量为0.850 0 mg.mL-1。结论对于成分简单的样品,采用二硝基水杨酸法测定玉米须保健饮料中总多糖含量,快速准确,稳定性好,可作为该制剂总多糖含量测定方法。

Quantitative determination of polysaccharides in traditional Chinese medicine health care beverage of stigma maydis by DNS method

Objective To find a best way to prepare the sample solution and establish the method for determining the content of polysaccharides in health care beverage of stigma maydis.Methods The best way to prepare the sample solution was found by orthogonal test,and the content of polysaccharides was measured by the 3,5-dinitrosalicylic acid（DNS） colorimetry,using glucose as the standard.Results The calibration curve was linear within the range of 8～56 μg·mL-1 for glucose.The average recovery was 101.83% with RSD=1.84%.The average content of polysaccharides was 0.850 0 mg·mL-1.Conclusion For the sample which composition wasn＇t complicated,it was fast,accurate and stable of DNS method to determine the content of polysaccharides.The method can be applied in determination of polysaccharides in health care beverage.
