慢性再生障碍性贫血患者血浆可溶性Fas,FasL水平与细胞凋亡的相关性研究

摘    要:目的 研究慢性再生障碍性贫血 (CAA)血浆可溶性Fas ,FasL(sFas ,sFasL)水平与细胞凋亡的相关性 ,探讨CAA的发病机制。方法 随机选择经确诊的CAA患者 39例 ,采用酶联免疫吸附双抗体夹心 (ELISA)法检测细胞凋亡及sFas ,sFasL水平。结果 CAA患者血浆sFas水平明显低于对照组 (P <0 .0 1) ,而细胞凋亡指数明显高于对照组 (P <0 .0 1)。二者变化水平呈负相关。血浆sFasL水平与对照组比较 ,差异无显著性 (P >0 .0 5 )。结论 CAA患者血浆sFas水平显著降低可导致造血细胞的过度凋亡 ;CAA发病过程中不但存在凋亡因素的增多 ,而且存在抗凋亡因素的减少

Abstract:Objective To investigate the relationship between plasma sFas,sFasL levels and apoptosis in chronic aplastic anemia (CAA) and to explore the pathogenesis of CAA.Methods 39 CAA patients were randomly selected.The apoptosis index and the levels of sFas,sFasL were assayed by ELISA technique.Results The sFas level in CAA was significantly lower than that in normal control group (P<0.01),but the apoptosis index in CAA was obviously higher than that in normal control group (P<0.01).The level of sFas was negatively correlated with apoptosis index.No significant difference was shown between sFasL level in CAA and in control group(P<0.05).Conclusion The decrease of plasma sFas level in CAA results in hematopoietic stem cell apoptosis.In the pathogenesis of CAA,factors which induce apoptosis increase and those which inhibit apoptosis decrease.
