损伤与正常脊髓蛋白质组的差异分析

摘    要:目的从整体水平探究损伤与正常脊髓在蛋白质组表达上的差异,并寻找两者间的差示蛋白。方法以横断损伤和正常大鼠脊髓为研究对象,应用三氯乙酸/丙酮法制备蛋白样品,经双向电泳和银染,分别获取大鼠损伤和正常脊髓的蛋白质组双向电泳银染图谱,通过图像扫描及图像分析,建立损伤与正常脊髓蛋白质组的匹配差示图谱,分析两者在蛋白质组表达上的变化。结果损伤脊髓蛋白质组图谱经图像分析后,可检测到746个蛋白点,正常脊髓蛋白质组图谱可检测到847个蛋白点;蛋白质组匹配差示图谱的分析发现,两者间存在411个差示蛋白,即:损伤脊髓蛋白质组中有155个蛋白点在正常脊髓蛋白质组中未找到相应的匹配蛋白点;同时,正常脊髓蛋白质组中也有256个蛋白点在损伤脊髓蛋白质组中未找到相应的匹配蛋白点。结论损伤与正常脊髓的蛋白质组存在明显差异。

Abstract:Objective To explore the differences of proteome between injured and normal rat spinal cord and find the differentially displayed proteins.Methods Trichloroacetic acid/acetone method was used to prepare the spinal cord tissue protein sample, and proteome maps of injured and normal rat spinal cord were obtained by 2-DE respectively. Proteome maps were digitized with the ImageScanner. The images analyses and establishment of the matching differentially displayed map between injured and normal rat spinal cord were performed with ImageMaster 2D Elite 3.01 software.Results After the analyses, 746 protein spots were distinguishable in proteome map of injured rat spinal cord and 847 protein spots in proteome map of normal rat spinal cord. The analysis of match differentially displayed map revealed that there were 411 differentially displayed proteins. That was, there were 155 differentially displayed proteins which were unique in the proteome of injured rat spinal cord and 256 differentially displayed proteins which were unique in the proteome of normal rat spinal cord.Conclusion There were significant difference in proteome between injured and normal rat spinal cord.
