IVIG对新生儿脐带血单核吞噬细胞表面分子的影响

摘    要:为从脐带血单核吞噬细胞(Mo/Mφ)表面CD14、HLA-DR和CD86分子表达的角度来探讨IVIG对新生儿免疫抑制作用,将IVIG、GM-CSF和LPS不同组合对Mo/Mφ进行刺激培养,利用双色免疫荧光抗体标记-流式细胞技术检测Mo/Mφ表面CD14、HLA-DR和CD86分子的表达。加入IVIG处理培养的脐带血Mo/Mφ表面CD14、HLA-DR和CD86分子的阳性细胞比率和相对荧光密度均明显降低,同时CD14+HLA-DR+和CD14+CD86+细胞的比率明显降低。IVIG可通过抑制脐带血Mo/Mφ表面CD14、HLA-DR和CD86分子的表达而抑制其抗原提呈能力,这可能是IVIG免疫抑制的机理之一。

Abstract:In order to investigate the inhibitory mechanism of intravenous immunoglobulin(IVIG) upon the surface molecules(CD14,HLA-DR,CD86) on neonatal umbilical cord blood monocytes/macrophages(Mo/Mφ),this kind of Mo/Mφ was stimulated with different combinations of IVIG,GM-CSF and LPS,and the expressions of CD14,HLA-DR and CD86 on Mo/Mφ were detected by means of dual-color immunofluorescence antibody staining and flow cytometry(FCM).In addition,the results were compared with those of the adults.It was found that the per-centages of CD14 +,HLA-DR + and CD86 + cells and the relative intensity of fluorescence were significantly re-duced when the Mo/Mφ were treated with IVIG.Also,IVIG could inhibit the activation of the Mo/Mφ induced by GM-CSF and LPS.It is concluded that IVIG can suppress the capacity of antigen presentation of umbilical cord blood Mo/Mφ,and this may be the mechanism for the inhibitory effect of intravenous immunoglobulin.
