| 摘 要： | 目的:探讨MLC2与TRE17之间的相互作用。方法:构建了MLC2原核表达质粒,在用pGEX蛋白表达系统表达和纯化GST-MLC2和GST蛋白后,用GST Pulldwon方法进一步观察MLC2与TRE17之间的相互作用。结果:限制性内切酶分析和DNA序列测定表明MLC2 cDNA完整ORF以正确的阅读框架插入pGEX-6P-1载体;IPTG诱导表达的GST-MLC2和GST蛋白纯度在90%以上;GST Pulldown分析表明癌蛋白TRE17(447)结合GST-MLC2融合蛋白,而不是对照GST蛋白。结论:MLC2是TRE17的特异性结合蛋白。

| Abstract: | Objective: To identify interaction of cytoskeletal MLC2 with oncogene TRE17.Methods: Plasmid GST-MLC2/pGEX was constructed in this study and used to express GST-MLC2 fusion protein.After purification of GST-MLC2 and GST proteins,MLC2 interaction with TRE17 was tested by GST pulldown assay.Results: The MLC2 cDNA was inserted into pGEX-6P-1 vector with correct open reading frame.After expression and purification of GST-MLC2 and GST proteins,GST pulldown assay suggested the association of TRE17(447) with MLC2,not GST protein.Conclusion: Skeletal MLC2 might be a binding partner of TRE17 oncoprotein. |
