染锰大鼠血有核细胞线粒体功能的体外及体内研究

摘    要:目的 探讨锰暴露对血有核细胞线粒体功能的影响。方法 利用大鼠血有核细胞线粒体进行体外、体内研究 ,观察锰染毒对线粒体呼吸功能、ATP生成及线粒体膜电位的影响。结果 体外研究表明 ,在不同浓度MnCl2 ·4H2 O (0 ,0 5 ,1 ,1 5 ,2 ,2 5mmol/L)与线粒体共同孵育 1 5min后 ,反映线粒体呼吸功能的呼吸控制比 (RCR)、ATP生成随着锰浓度的增加而逐渐降低 ,其中 2 5mmol/L剂量组与对照组相比差异有显著性 ,且 2 5mmol/L锰可致线粒体的膜电位降低 ;接触锰 2 5mmol/L 1 0min后线粒体功能开始下降 ,1 5min后RCR、ATP生成、膜电位降低与对照组相比 ,差异有显著性。体内研究表明 ,在大鼠均未见神经行为方面有中毒表现时 ,高剂量组 (2 5mg/kg)RCR、ATP生存均显著降低 ,其余各组与对照组相比 ,差异无显著性。结论 锰对大鼠血有核细胞线粒体功能具有损害作用 ,血有核细胞线粒体功能异常可能有助于锰中毒的早期发现和诊断

Abstract:Objective To study effects of Mn on rat hemakaryocyte mitochondrial function.Methods In vivo and in vitro studies,mitochondrial respiratory function(RCR),ATP production and mitochondrial membrane potential are monitored.Results In vitro study,it showed that after Mn exposure of different concentration(0,0^5,1,1^5,2,2^5 mmol/L)for 15 min,RCR and ATP production gradually reduced with the elevation of Mn concentration,in which the group of 2^5 mmol/L Mn was significantly different compared with the control group,at the same time,causing the drop of mitochondrial membrane potential.In addition,2^5 mmol/L group with different exposure time indicated that after 10 min exposure mitochondrial function began to drop,and 15 min later,RCR,ATP production and mitochondrial membrane potential showed significantly different compared with the control group,In vivo study,it showed that in high dose group(25 mg/kg)both RCR and ATP decreased markedly.The rest groups had no significant difference as compared with the control group.Conclusions Mn exposure could exert adverse effect on rat hemokaryocyte mitochondrial function.
