舒芬太尼后处理对大鼠肠缺血再灌注时肝组织损伤的影响

摘    要:目的研究舒芬太尼后处理对大鼠肠缺血再灌注时肝组织损伤的影响。方法成年Wista大鼠40只,随机分为5组(n=8):假手术组(S组)、肠缺血再灌注组(IIR组)、舒芬太尼后处理低、中、高剂量组(SP1、SP2、SP3组)。于再灌注3h时处死大鼠,取肝组织,用免疫组织化学方法检测Bcl-2与Bax表达水平,用TUNEL法检测并计算肝细胞凋亡指数(AI)。结果与S组相比,IIR组和SP1-3组Bcl-2与Bax表达上调,肝细胞AI升高,IIR组Bcl-2/Bax比值降低,SP1-3组Bcl-2/Bax比值升高(P〈0.05);与IIR组相比,SP1-3组Bcl-2表达、Bcl-2/Bax比值升高,Bax表达下调,肝细胞AI降低(P〈0.05)。结论舒芬太尼后处理可抑制大鼠肠缺血再灌注时肝组织损伤,其机制可能与上调Bcl-2表达及抑制Bax上调表达有关。

Abstract:Objective To investigate the effects of sufentanil postconditioning liver tissue injury induced by intestinal ischemiareperfusion in rats.Methods Forty Wistar rats were randomly divided into 5 groups(n=8):Shamoperation group(S group),intestinal ischemia reperfusion group(IIR group) and postconditioning of low-dosage sufentanil group(SP1 group)、middle-dosage group(SP2 group) and high-dosage group(SP3 group).After reperfusion two hours,Immunohistochemistry was used to detect the Bcl-2 and Bax protein of hepatic tissue.The presence of apoptosis was detected by TUNEL.Results Compared with the sham-operated group,express-ion of Bax,Bcl-2 and apoptosis index of hepatic cell increased in intestinal ischemia reperfusion group and sufentanil postconditioning group.Bcl-2/Bax ratio is lower in intestinal ischemi-a-reperfusion group.Bcl-2/Bax ratio is higher in sufentanil postconditioning group(P0.05).Compared with the intestinal ischemia reperfusion group,expression of Bcl-2 and Bcl-2/Bax ratio is higher and expression of Bax and apoptosis index of hepatic cell is lower in sufentanil postconditioning group(P0.05).Conclusion Sufentanil postconditioning can protect liver injury induced by intestinal ischemia-reperfusion in rats.One of mechanisms of protection is may be that Sufentanil increase the expression of Bcl-2 protein and decrease the expression of Bax protein.
