| 摘 要： | 目的探讨ERK1/2抑制剂PD98059对人子宫颈癌细胞株Siha细胞增殖和凋亡的影响。方法不同浓度（10μmol/L25μmol/L,50μmol/L,100μmol/L）PD98059作用于Siha,取不同时间点（6h,12h,24h,48h）,然后采用MTT法检测细胞的增殖情况,流式细胞仪检测细胞的凋亡情况。结果 PD98059能够抑制Siha细胞增殖,诱导细胞凋亡,并且呈时间-剂量依赖性（P〈0.05）。结论 PD98059能够抑制子宫颈癌细胞株Siha细胞增殖,诱导凋亡,可能与PD98059抑制了RAS-RAF-MEK-ERK信号转导途径有关。

| Abstract: | Objective To study the effect of ERK1/2 Inhibitor PD98059 on proliferation and Apoptosis of human cervical cancer cell lines Siha.Methods The Siha cells were treated by PD98059（PD98059 5μmol/L,10μmol/L,25μmol/L,50μmol/L） for 6h,12h,24h and 48h,MTT assay detects cell proliferation,flow cytometry evaluate cell Apoptosis.Results The proliferation and Apoptosis of Siha cells was inhibited in some time and dose dependent patterns by PD98059（P0.05）.Conclusions PD98059 can inhibit the proliferation and induce Apoptosis of human cervical cancer cell lines Siha,It have to do with PD98059 may inhibit the RAS-RAF-MEK-ERK signal transduction pathway. |
