| 摘 要： | 目的：探讨低氧预适应（HP）对大鼠自体肝移植后肠道TLR4蛋白表达的影响。方法：将SD大鼠随机分为正常对照组（NC）、自体肝移植（AT）组和低氧预适应＋AT（HP）组3组,每组24只,采用改良的自体肝移植模型。HP组术前用8%氮氧混合气体处理90 min,分别于术后1、6、12 h处死大鼠取小肠标本,抽血检测血清ALT及AST水平,采用免疫组化法测定各组小肠组织TLR4蛋白表达情况,及应用逆转录-聚合酶反应（RT-PCR）检测小肠组织TNF-αmRNA的含量。结果：HP组血清ALT、AST水平,小肠组织TLR4蛋白表达及TNF-αmRNA的含量均显著低于AT组,但均高于NC组,差异均有统计学意义（P〈0.05）。结论：HP通过下调肝移植大鼠小肠组织TLR4蛋白的表达,进一步抑制TLR4信号转导通路,减少TNF-α等细胞因子的合成与释放,从而减轻对移植肝功能的损伤。
