摘    要:目的建立反相高效液相色谱法同时测定黄芩药材及其制剂中黄芩苷(1)、汉黄芩苷(2)、黄芩素(3)和汉黄芩素(4)的含量。方法采用Agilent Zorbax SB-C18色谱柱(4.6 mm×250 mm,5μm);以乙腈-0.15%三氟乙酸水为流动相,梯度洗脱;流速:1.0 ml·min^-1;检测波长:280 nm;柱温:室温。结果已测定的4种黄酮在以下浓度范围内呈现良好的线性:(1)0.51~164 mg·L^-1(r=0.9999),(2)0.33~104 mg·L^-1(r=0.9998),(3)0.24~76 mg·L^-1(r=0.999 8),(4)0.31~100mg·L^-1(r=0.9999)。平均回收率(n=3)分别为:(1)98.47%(RSD=1.35%),(2)100.73%(RSD=0.57%),(3)101%(RSD=2.63%),(4)100.77%(RSD=0.27%)。结论该方法简便,准确,灵敏,结果可靠,可用于黄芩药材及其制剂的质量控制。

Abstract:Aim To establish a RP-HPLC method for simultaneous determination of baicalin(1),wogonoside(2),baicalein(3)and wogonin(4)in Radix Scutellariae and its related traditional Chinese medicinal preparations.Methods These four flavonoids were successfully analyzed on an Agilent Zorbax SB-C18 reversed-phase chromatographic column(4.6 mm × 250 mm,5 μm) by gradient elution using 0.15%(v/v) aqueous trifluoroacetic acid and acetonitrile as the mobile phase at room temperature.The flow rate was 1 ml·min^-1,and detection wavelength was set at 280 nm.Results The method showed good linearity in the ranges of(1) 0.51~164 mg·L^-1(r=0.999 9),(2) 0.33~104 mg·L^-1(r=0.999 8),(3) 0.24~ 76 mg·L^-1(r=0.999 8),(4) 0.31~100 mg·L^-1(r=0.999 9) for the four flavonoids respectively;and the mean recovery(n=3) of the four invested compounds were(1) 98.47%(RSD =1.35%),(2)100.73%(RSD=0.57%),(3) 101%(RSD=2.63%),(4)100.77%(RSD=0.27%).Conclusion The developed new method is simple,precise,accurate,and reliable for quality evaluation of Radix Scutellariae and its related traditional Chinese medicinal preparations.
