骨形成蛋白-2对人牙乳头细胞内Smad1蛋白表达量的影响

摘    要:目的:观察人牙乳头细胞内Smad1蛋白的表达及骨形成蛋白-2(bone morphogenetic protein-2,BMP2)对人牙乳头细胞内Smad1蛋白表达量影响,方法:原代培养的人牙乳头细胞用BMP2处理6h,12h,24h,48h后,提取总蛋白质,采用Western Bolt法,从翻译水平观察Smad1基因的表达变化。结果:从翻译水平观察到Smad1基因在人牙乳头细胞内的表达,但在BMP2作用48h内,Smad1蛋白表达量无显著变化,结论:人牙乳头细胞内存在Smad1信号转导途径,牙乳头细胞内Smad1蛋白表达量不受MBP2影响。

Abstract:Objective To investigate the expression of Smad1 gene and regulation by bone morphogenetic protein-2(BMP2)in the human dental papilla cells (HDPC).Methods Treatment by BMP2 after 6h,12h,24h and 48h,endogenous Smad1 protein expression and modulation by BMP2 in the cultured HDPC were evaluated by Western Blot analyses.Results Cultured HDPC expressed Smad1 protein on translation level.The expression of Smad1 expression wasn't significantly altered by BMP2 within 48h treatment.Conclusion Smad1 signaling is present in the HDPC,but the expression of Smad1 protein is not influenced by BMP2.
